Gastrin, is a hormone that normally formed by mucosal cells in the gastric antrum and by the D cells of the pancreatic islets. Its primary function is to stimulate secretion of HCl by the gastric mucosa. HCl, in turn, inhibits gastrin formation. It also responsible for stimulating smooth muscle contraction and increasing blood circulation and water secretion in the stomach and intestine. Gastrin is regulated by epidermal growth factor in both mice and humans. Gastrin is excreted in excess by pancreatic tumors in the Zollinger-Ellison syndrome. Gastrin-Releasing Peptide (GRP) stimulates the release of gastrin as well as other gastrointestinal hormones and also acts as an autocrine growth factor for certain cell types. High levels of GRP are found in the human lung just after birth and levels decrease thereafter in parallel with the observed disease in a number of pulmonary neuroendocrine cells. This antibody recognizes a glycoprotein of 75 kDa, identified as low affinity Nerve Growth Factor (NGF) Receptor (p75NGFR) or Neurotrophin Receptor (p75NTR). NGFR is expressed in various neural crest cells and their tumors such as melanocytes, melanomas, neuroblastomas, pheochromocytomas and neurofibromas. Reportedly, anti-NGFR is a reliable marker for desmoplastic and neurotropic melanomas. NGFR is expressed in mature non-neural cells such as perivascular cells, dental pulp cells, lymphoidal follicular dendritic cells, basal epithelium of oral mucosa and hair follicles, prostate basal cells, and myoepithelial cells. Expression of NKX2.2 has been found in neuroendocrine tumors of the gut, making it a potential marker for the study of gastrointestinal neuroendocrine tumors. More recently, NKX2.2 protein was identified as a target of EWS-FLI-1, the fusion protein specific to Ewing sarcoma, and was shown to be differentially upregulated in Ewing sarcoma on the basis of array-based gene expression analysis. It acts as a valuable marker for Ewing sarcoma, with a sensitivity of 93% and a specificity of 89%, and aids in the differential diagnosis of small round cell tumors.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. SOX2 is a member of the SRY-related HMG-box (SOX) family of transcription factors involved in the regulation of embryonic development and in the determination of cell fate. At present, 30 Sox genes have been identified. SOX2 is required for stem cell maintenance in the central nervous system, and it also regulates gene expression in the stomach. SOX2 is necessary for regulating multiple transcription factors that affect Oct 3/4 expression. Interleukin 10 (IL10) is a cytokine produced primarily by monocytes and to a lesser extent by lymphocytes. This cytokine has pleiotropic effects in immuno-regulation and inflammation. It down-regulates the expression of Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice suggested the function of this cytokine as an essential immuno-regulator in the intestinal tract. EBAG9, also known as RCAS1, is an estrogen-transcribed protein. Soluble and membranous RCAS1 proteins may play a role in the immune escape of tumor cells by promoting T lymphocyte inhibition of growth and apoptosis. RCAS1 is expressed in a wide variety of cancers, including uterine, ovarian, and lung cancer cells, and acts as a ligand for a putative receptor present on peripheral lymphocytes. RCAS1 is highly expressed not only in cancer cells but also in non-tumor bile duct cells subject to immune attack. RCAS1 inhibits the in vitro growth of receptor-expressing cells and induces apoptosis, contributing to the ability of tumor cells to evade host immune surveillance. Recognizes a protein of 75 kDa, identified as mu heavy chain of human immunoglobulins. It does not cross-react with alpha (IgA), gamma (IgG), epsilon (IgE), or delta (IgD), heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. Monomeric IgM is expressed as a membrane bound antibody on the surface of B cells and as a pentamer when secreted by plasma cells. Cytokeratin 17 (CK17) is normally expressed in the basal cells of complex epithelia but not in stratified or simple epithelia. Antibody to CK17 is an excellent tool to distinguish myoepithelial cells from luminal epithelium of various glands such as mammary, sweat and salivary. CK17 is expressed in epithelial cells of various origins, such as bronchial epithelial cells and skin appendages. It may be considered as epithelial stem cell marker because CK17 Ab marks basal cell differentiation. CK17 is expressed in SCLC much higher than in LADC.
