Biotium MSH6 (DNA Mismatch Repair Protein)(MSH6/3086), 0.2mg/mL
The finding that mutations in DNA mismatch repair genes are associated with hereditary nonpolyposis colorectal cancer (HNPCC) has resulted in considerable interest in the understanding of the mechanism of DNA mismatch repair. Initially, inherited mutations in the MSH2 and MLH1 homologs of the bacterial DNA mismatch repair genes mutS and mutL were demonstrated at high frequency in HNPCC and were shown to be associated with microsatellite instability. A member of the mismatch repair family, GTBP (also designated MSH6), is an MSH2-related protein that binds to DNA containing G/T mismatches. Findings suggest that the mismatch-binding factor in human cells is composed of a heterodimer of GTBP and MSH2. Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. CF Dyes offer exceptional brightness and photostability. Note: Conjugates of blue fluorescent dyes like CF405S and CF405M are not recommended for detecting low abundance targ

Biotium CD63 (Late Endosomes Marker)(LAMP3/2789), 0.2mg/mL
This MAb recognizes protein of 26 kDa-60 kDa, which is identified as CD63. Its epitope is different from that of MAb LAMP3/529. The tetraspanins are integral membrane proteins expressed on cell surface and granular membranes of hematopoietic cells and are components of multi-molecular complexes with specific integrins. The tetraspanin CD63 is a lysosomal membrane glycoprotein that translocates to the plasma membrane after platelet activation. CD63 is expressed on activated platelets, monocytes and macrophages, and is weakly expressed on granulocytes, T cell and B cells. It is located on the basophilic granule membranes and on the plasma membranes of lymphocytes and granulocytes. CD63 is a member of the TM4 superfamily of leukocyte glycoproteins that includes CD9, CD37 and CD53, which contain four transmembrane regions. CD63 may play a role in phagocytic and intracellular lysosome-phagosome fusion events. CD63 deficiency is associated with Hermansky-Pudlak syndrome and is stro

Biotium Prohibitin (Mitochondrial Marker)(PHB/3230), 1mg/mL
Recognizes a protein of 30 kDa which is identified as Prohibitin, an evolutionarily conserved protein with homologues found in yeast to man. It is located in the inner membrane of mitochondria. Although prohibitin mRNA and protein expression occurs throughout the cell cycle, maximum levels are detected during the G1/S phase transition and minimum levels are seen in S phase and the G2/mitosis boundary. Prohibitin is located exclusively in the mitochondria with the highest concentration on the inner membrane. Prohibitin is an ideal mitochondrial marker. It shows antiproliferative activity and has been proposed to play a role in normal cell cycle regulation, replicative senescence, cellular immortalization, and tumor suppression. Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. CF Dyes offer exceptional brightness and photostability. Note: Conjugates of blue fluorescent dyes like CF405S and CF405M are not recommended fo

Biotium Elastin (ELN) (Marker of Arterial Stiffness and Atherosclerosis)(ELN/3131R), CF647 conjugate, 0.1mg/mL
Elastin is a polymeric protein and is the main component of the extracellular matrix of arteries. It is synthesized and secreted as a soluble, single-chain protein (tropoelastin), which undergoes a number of post-translational modifications prior to its organization into an elastic fiber in the extracellular space. Elastin performs a regulatory function during arterial development by controlling proliferation of smooth muscle and stabilizing arterial structure. It imparts elasticity to the connective tissue. During aging, the elasticity of connective tissue is reduced because of the cross-linking of collagenous fibers with elastin. The abnormal accumulation of elastic tissue in blood vessels is found in atherosclerosis and hypertension. Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. CF Dyes offer exceptional brightness and photostability. Note: Conjugates of blue fluorescent dyes like CF405S and CF405M are not reco

Biotium NKX3.1 (Metastatic Prostate Adenocarcinoma Marker)(NKX3.1/3350), CF740 conjugate, 0.1mg/mL
NKX3. 1 is a prostate specific gene encoding a transcription factor that plays an important role in normal prostate development and carcinogenesis. It is a prostatic tumor suppressor gene located on chromosome 8p21. 2, which frequently undergoes a loss of heterozygosity. NKX3. 1 expression is highly restricted in prostate epithelial cells and therefore can be used as a diagnostic biomarker for prostate cancer and other metastatic lesions of prostatic origin. Furthermore, NKX3. 1 shows better sensitivity than Prostate Specific Antigen (PSA) for identifying metastatic prostatic adenocarcinoma. This suggests that immunohistochemical staining of NKX3. 1, along with other prostate-restricted markers, may be valuable for the definitive determination of prostatic origin in poorly differentiated metastatic carcinomas. Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. CF Dyes offer exceptional brightness and photostability. Note

Biotium HLA-Pan (MHC II)(rHLA-Pan/3475), CF568 conjugate, 0.1mg/mL
Reacts with a common epitope of human major histocompatibility (MHC) class II antigens, HLA-DP, -DQ and -DR. Human MHC class II antigens are transmembrane glycoproteins composed of an alpha chain (36 kDa) and a beta chain (27 kDa). They are expressed primarily on antigen presenting cells such as B lymphocytes, monocytes, macrophages, and thymic epithelial cells and are also present on activated T lymphocytes. Human MHC class II genes are located in the HLA-D region that encodes at least six alpha and ten beta chain genes. Three loci, DR, DQ and DP, encode the major expressed products of the human class II region. The human MHC class II molecules bind intracellularly processed peptides and present them to T-helper cells. They, therefore, have a critical role in the initiation of the immune response. Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. CF Dyes offer exceptional brightness and photostability. Note: Conjugates

Biotium MUC3 (Mucin 3)(MUC3/2992R), CF568 conjugate, 0.1mg/mL
Recognizes a protein of HMW, identified as mucin 3 glycoprotein (MUC3). Its epitope localizes between aa SITTTE. This MAb shows no cross-reaction with human milk fat globule membranes, MUC1, or MUC2. MUC3 is distributed in colon and rectum, and is also present to a lesser extent in breast, lung and salivary gland tissues. The Mucins are a family of highly glycosylated, secreted proteins with a basic structure consisting of a variable number of tandem repeats (VNTRs) encoded by 60 base pairs (Mucin 1), 69 base pairs (Mucin 2) and 51 base pairs (Mucin 3). The number of repeats is highly polymorphic and varies among different alleles. Mucin 1 proteins are expressed as type I membrane proteins in addition to secreted forms. Mucin 1 is aberrantly expressed in epithelial tumors including breast carcinomas. Mucin 2 coats the epithelia of the intestines and airways and is associated with colonic tumors. Mucin 3 is a major component of various mucus gels and is broadly expressed in no

Biotium Cytochrome C (Mitochondrial Marker)(CYCS/3128R), CF568 conjugate, 0.1mg/mL
Recognizes an epitope within amino acids 93-104 of pigeon Cytochrome C, a well-characterized mobile electron transport protein that is essential to energy conversion in all aerobic organisms. In mammalian cells, this highly conserved protein is normally localized to the mitochondrial inter-membrane space. More recent studies have identified cytosolic cytochrome c as a factor necessary for activation of apoptosis. During apoptosis, cytochrome c is trans-located from the mitochondrial membrane to the cytosol, where it is required for activation of caspase-3 (CPP32). Overexpression of Bcl-2 has been shown to prevent the translocation of cytochrome c, thereby blocking the apoptotic process. Overexpression of Bax has been shown to induce the release of cytochrome c and to induce cell death. The release of cytochrome c from the mitochondria is thought to trigger an apoptotic cascade, whereby Apaf-1 binds to Apaf-3 (caspase-9) in a cytochrome c-dependent manner, leading to caspase-9

Biotium Alpha Actinin 4 / ACTN4(93), CF568 conjugate, 0.1mg/mL
Alpha actinins belong to the spectrin gene superfamily which represents a diverse group of cytoskeletal proteins, including the alpha and beta spectrins and dystrophins. There are four tissue-specific -actinins, namely -actinin-1, -actinin-2, -actinin-3 and -actinin-4, which are localized to muscle and non-muscle cells, including skeletal, cardiac and smooth muscle cells, as well as within the cytoskeleton. Alpha actinin is an actin-binding protein with multiple roles in different cell types. In nonmuscle cells, the cytoskeletal isoform is found along microfilament bundles and adherens-type junctions, where it is involved in binding actin to the membrane. In contrast, skeletal, cardiac, and smooth muscle isoforms are localized to the Z-disc and analogous dense bodies, where they help anchor the myofibrillar actin filaments. This gene encodes a nonmuscle, alpha actinin isoform which is concentrated in the cytoplasm, and thought to be involved in metastatic processes. Each -act

Biotium Desmoglein-3 (Squamous Cell Marker)(DSG3/2796), CF568 conjugate, 0.1mg/mL
Recognizes a protein of 130 kDa, identified as Desmoglein-3 (DSG3). This MAb is highly specific to Desmoglein-3 and does not cross-react with other members of the Desmoglein-family. DSG3 is a calcium-binding transmembrane glycoprotein component of desmosomes in vertebrate epithelial cells. Research has shown that DSG3 has a very high sensitivity (80%) and specificity (100%) in recognizing squamous cell carcinoma (SqCC). Therefore, DSG3 is considered a very important marker for lung SqCC and can be a useful ancillary marker to separate SqCC from other subtypes of lung cancer. Moreover, studies have shown that DSG3 expression in lung SqCC may indicate a poor prognosis. Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. CF Dyes offer exceptional brightness and photostability. Biotium Cytokeratin 15 (Esophageal Squamous Cell Carcinoma Marker)(KRT15/2957), 0.2mg/mL
Keratin 15 is a type I keratin which is expressed only in basal keratinocytes in stratified epithelia and does not appear to have a natural type II expression partner. Keratin 15 is down regulated in activated keratinocytes. Cytokeratin 15 is a specific marker of stem cells of the hair-follicle bulge and may be a useful marker for diagnosis between basal cell carcinoma (BCC) and trichoepithelioma. Trichoblastoma are benign neoplasms of follicular differentiation frequently found in nevus sebaceous. Many morphologic features are shared with nodular basal cell carcinoma, sometimes rendering a diagnosis difficult. Trichoblastoma and BCC show variable expression of Cytokeratin 15 and Cytokeratin 19, and absence of hair keratins. Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. CF Dyes offer exceptional brightness and photostability. Biotium XRCC3(10F1/6), CF568 conjugate, 0.1mg/mL
The x-ray repair cross-complementing (XRCC) proteins are responsible for efficiently repairing and maintaining genetic stability following DNA base damage. These genes share sequence similarity with the yeast DNA repair protein Rad51. XRCC1 is a protein that facilitates the DNA base excision repair pathway by interacting with DNA ligase III and DNA polymerase to repair DNA single-strand breaks. XRCC2 and XRCC3 are both involved in maintaining chromosome stability during cell division. XRCC2 is required for efficient repair of DNA double-strand breaks by homologous recombination between sister chromatids, and XRCC3 interacts directly with Rad51 to cooperate with Rad51 during recombinational repair. XRCC4 is an accessory factor of DNA Ligase IV that preferentially binds DNA with nicks or broken ends. XRCC4 binds to DNA Ligase IV and enhances its joining activity, and it is also involved in V(D)J recombination. Any defect in one of the known components of the DNA repair/V(D)J re

Biotium CD11c (Dendritic Cell Marker) (ITGAX/1284), CF555 conjugate, 0.1mg/mL
This antibody recognizes a protein of 145 kDa, identified as CD11c. CD11c (ITGAX), a member of the leukointegrin family, shares the same beta subunit with other members of the leukocyte adhesion molecule family, which includes CD11a (LFA-1), CD11b (MAC-1) and CD11d (ITGAD), but has a unique alpha chain. CD11c has been shown to play a role in phagocytosis, cell migration, and cytokine production by monocytes/macrophages as well as induction of T-cell proliferation by Langerhans cells. CD11c is expressed prominently on the plasma membranes of monocytes, tissue macrophages, NK cells, and most dendritic cells (DCs). A lower level of expression is also observed on neutrophils as a result of its high level of expression on most DCs. An antibody to CD11c may aid in identification of lesions with histiocytic origin. It may also been used as a marker for hairy cell leukemia in paraffin-embedded tissues.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes

Biotium Lymphocyte Specific Protein 1 (LSP1) / pp52(LSP1/3025), CF740 conjugate, 0.1mg/mL
LSP1 is a hematopoietic-expressed gene that encodes an F-actin-binding, leukocyte-specific (including B and T lymphocytes, granulocytes and macrophages), phosphoprotein. However, mRNA splice variants that do not encode the lympho-specific protein are expressed from this gene in nonlymphoid cell lines as well (myocytes, stromal cells and fibroblasts), suggesting pp52 has a divergent role in signal transduction. The pp52 (LSP1) locus maps to human chromosome 11p15. 5, which is implicated in tumor-related chromosomal translocations found in chronic lymphocytic leukemia. The pp52 promoter contains key elements that control transcriptional activity including an initiator specifying the unique 5' terminus of pp52 mRNA, tandem pairs of Ets and SP1 motifs, and a single C/EBP motif. LSP1 binds the cytoskeleton and has been implicated in affecting cytoskeletal remodeling in a variety of leukocyte functions, including cell motility and chemotaxis.
