Besides its role as a glycolytic enzyme, mammalian GPI can function as a tumor-secreted cytokine and an angiogenic factor (AMF) that stimulates endothelial cell motility. EBAG9, also known as RCAS1, is an estrogen-transcribed protein. Soluble and membranous RCAS1 proteins may play a role in the immune escape of tumor cells by promoting T lymphocyte inhibition of growth and apoptosis. RCAS1 is expressed in a wide variety of cancers, including uterine, ovarian, and lung cancer cells, and acts as a ligand for a putative receptor present on peripheral lymphocytes. RCAS1 is highly expressed not only in cancer cells but also in non-tumor bile duct cells subject to immune attack. RCAS1 inhibits the in vitro growth of receptor-expressing cells and induces apoptosis, contributing to the ability of tumor cells to evade host immune surveillance. High expression of RCAS1 significantly correlates with tumor progression and with poor outcome for many cancer patients. The soluble sulfotransferases contribute to the elimination of xenobiotics, the activation of procarcinogens and the regulation of hormones by catalyzing the sulfate conjugation of these substances. SULT1E1, also known as STE, is a 294 amino acid member of the SULT1 family. Localized to the cytoplasm and expressed in intestine, liver and kidney, SULT1E1 exists as a homodimer that is thought to control estrogen receptor (ER) levels by sulfurylating free estradiol. Defects in the gene encoding SULT1E1 are associated with an increased risk for endometrial cancer, suggesting a role for SULT1E1 in tumorigenesis. CD9 is a type IV transmembrane glycoprotein with four transmembrane domains. CD9 on pre-B cells may play a role in cell-cell adhesion. CD9 is expressed on early B cells, eosinophils, basophils and activated T cells and is a major component of the platelet cell surface. Creatine kinases (CK) are a large family of isoenzymes that regulate levels of ATP in subcellular compartments, where they provide ATP at sites of fluctuating energy demand by the transfer of phosphates between creatine and adenine nucleotides. CKs provide the energy of phosphate hydrolysis necessary to drive the normal function of many cellular systems. In cells, the cytosolic CK enzymes consist of two subunits, which can be either B (brain type) or M (muscle type). There are three different isoenzymes: CKMM, CKBB and CKMB. Recognizes a cluster of proteins between 70-80 kDa, identified as tyrosinase. Occasionally a minor band at 55 kDa is also detected. This MAb shows no cross-reaction with MAGE-1 and tyrosinase-related protein 1, TRP-1/gp75. Tyrosinase is a copper-containing metalloglycoprotein that catalyzes several steps in the melanin pigment biosynthetic pathway; the hydroxylation of tyrosine to L-3, 4-dihydroxy-phenylalanine (dopa), and the subsequent oxidation of dopa to dopaquinone. Mutations of the tyrosinase gene occur in various forms of albinism. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. Staining of melanomas with this MAb shows tyrosinase in melanotic as well as amelanotic variants. This MAb is a useful marker for melanocytes and melanomas. ZAP70 is a 70 kDa protein tyrosine kinase found in T-cells and natural killer cells. Control of this protein translation is via the IgVH gene. ZAP70 protein is expressed in leukemic cells of approximately 25% of chronic lymphocytic leukemia (CLL) cases as well. Anti-ZAP70 expression is an excellent surrogate marker for the distinction between the Ig-mutated (anti-ZAP70 negative) and Ig-unmutated (anti-ZAP70 positive) CLL subtypes and can identify patient groups with divergent clinical courses. Calbindin 1, also known as calbindin D28K, is a member of a family of calcium binding proteins. This protein family also includes calmodulin, calbindin 2, S-100, and troponin C. Calbindin 1 is a 261-amino acid protein with 6 EF-hand domains, 4 of which are active calcium-binding domains. Expressed in brain, ovary, uterus, testis, pancreas, liver, kidney and intestine, calbindin 1 acts as a calcium-buffering agent and alters the activity of the plasma membrane ATPase. In neuronal cells, calbindin 1 modulates calcium channel activity, calcium transients and intrinsic neuronal firing activity. Myoglobin is a cytosolic oxygen-binding protein responsible for the storage and diffusion of oxygen within myocytes. Expression of myoglobin is highest in skeletal and cardiac muscle. Myoglobin is well accepted as an O2-storage protein in muscle, capable of releasing O2 during periods of hypoxia or anoxia. In combination with other striated muscle markers such as vimentin and myogenin, myoglobin is helpful in the identification of rhabdomyosarcoma and tumors with skeletal muscle differentiation. Reportedly, myoglobin is expressed on epithelial cancer cells due to changed metabolic and environmental conditions. This MAb recognizes a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. This antibody cocktail recognizes three melanoma-specific proteins, which include MART-1, Tyrosinase and gp100. MART-1 is a newly identified melanocyte differentiation antigen recognized by autologous cytotoxic T lymphocytes. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. Function of gp100 is not known but it is reported to be a useful marker for melanocytes and melanomas. This cocktail of three markers is designed for extremely sensitive labeling of formalin-fixed, paraffin-embedded melanomas and other tumors showing melanocytic differentiation.

Elastin is a polymeric protein and is the main component of the extracellular matrix of arteries. It is synthesized and secreted as a soluble, single-chain protein (tropoelastin), which undergoes a number of post-translational modifications prior to its organization into an elastic fiber in the extracellular space. Elastin performs a regulatory function during arterial development by controlling proliferation of smooth muscle and stabilizing arterial structure. It imparts elasticity to the connective tissue. During aging, the elasticity of connective tissue is reduced because of the cross-linking of collagenous fibers with elastin. The specificity of this monoclonal antibody to its intended target was validated by HuProt™Array, containing more than 19,000, full-length human proteins.The intestine-specific transcription factors CDX1 and CDX2 are important for directing intestinal development, differentiation, proliferation and maintenance of the intestinal phenotype. CDX2 protein expression has been seen in GI carcinomas. Anti-CDX2 has been useful to establish GI origin of metastatic adenocarcinomas and carcinoids and is especially useful to distinguish metastatic colorectal adenocarcinoma from lung adenocarcinoma. However, mucinous carcinomas of the ovary also express CDX2 protein.
