T-cell leukemia/Lymphoma Protein 1A (TCL-1A), also known as p14TCL1 is a product of the TCL1 gene that is involved in T-cell prolymphocytic leukemia (TPLL). T-PLL is a rare form of mature T-cell leukemia, which is consistently associated with chromosomal rearrangements characterized by the juxtaposition of the TCRA locus on chromosome 14q11 and the TCL1A gene on 14q32. 13. This antibody recognizes a protein of 35 kDa, which is identified as tartrate-resistant acid phosphatase (TRAcP). It exists as two isoforms (5a and 5b). This MAb reacts with both the isoforms. Serum TRAcP 5a is secreted by macrophages and dendritic cells and increased in many patients of rheumatoid arthritis.Serum TRAcP 5b is produced from osteoclasts and elevated during bone resorption. TRAcP is an iron containing glycoprotein, which catalyzes the conversion of orthophosphoric monoester to alcohol and orthophosphate. S100 belongs to the family of calcium binding proteins. S100A and S100B proteins are two members of the S100 family. S100A is composed of an alpha and a beta chain whereas S100B is composed of two beta chains. This antibody is specific against an epitope located on the beta-chain (i.e. in S-100A and S-100B) but not on the alpha-chain of S-100 (i.e. in S-100A and S100A0). This antibody can be used to localize S-100A and S-100B in various tissue sections. The human papilloma virus (HPV) family of DNA tumor viruses includes HPV16, a strain that is responsible for the largest number of cases of cervical cancers linked to the family. HPV16E1 and HPV16E2 are proteins that are involved in the regulation of viral DNA replication and are important for infected cell homeostasis. HPV16E2 specifically regulates the expression of the E6 and E7 oncoproteins by binding to four sites within the viral long control region, possibly involving interactions with nuclear hormone receptors. Integration of the HPV genome into the host DNA usually disrupts the HPV16E2 gene open reading frames, resulting in an overexpression of E6 and E7 genes, an event that may lead to the malignant transformation of cervical cancer. This antibody recognizes a protein of 104 kDa-110 kDa, characterized as major vault protein (MVP). Vaults are large ribonucleoprotein particles (RNPs) present in all eukaryotic cells. They have a complex morphology, including several small molecules of RNA, but a single protein species. The MVP accounts for >70% of their mass. Their shape is reminiscent of the nucleopore central plug. Treatment of cells with estradiol increases the amount of MVP in nuclear extract. The hormone-dependent interaction of vaults with ER is prevented in vitro by sodium molybdate. Antibodies to estrogen, progesterone and glucocorticoid receptors are able to co-immunoprecipitate the MVP. MVP is overexpressed in many neoplastic tissues and cell lines. This antibody recognizes a protein of HMW, identified as mucin 3 glycoprotein (MUC3). This MAb shows no cross-reaction with human milk fat globule membranes, MUC1, or MUC2. The Mucins are a family of highly glycosylated, secreted proteins with a basic structure consisting of a variable number of tandem repeats (VNTRs) encoded by 60 base pairs (Mucin 1), 69 base pairs (Mucin 2) and 51 base pairs (Mucin 3). The number of repeats is highly polymorphic and varies among different alleles. Mucin 1 proteins are expressed as type I membrane proteins in addition to secreted forms. Complement C1q A Chain (C1qa) is a subunit of C1q. C1q is composed of 6 polypeptide chains each of C1qa, C1qb, and C1qc. Keratin 15 is a type I keratin which is expressed only in basal keratinocytes in stratified epithelia and does not appear to have a natural type II expression partner. Keratin 15 is down regulated in activated keratinocytes. Cytokeratin 15 is a specific marker of stem cells of the hair-follicle bulge and may be a useful marker for diagnosis between basal cell carcinoma (BCC) and trichoepithelioma. Trichoblastoma are benign neoplasms of follicular differentiation frequently found in nevus sebaceous. This antibody recognizes a protein of ~150 kDa, identified as Desmoglein-1 (DSG1). Desmoglein-1 is a member of the desmosomal cadherin family. Desmosomes are intercellular adhering junctions that represent cell surface attachment sites for intermediate filament. Desmocollins and desmogleins are the main desmosomal transmembrane proteins. Desmogleins consist of Dsg1, Dsg2, Dsg3, and Dsg4 isoforms.
