CD45R, also designated CD45 and PTPRC, has been identified as a transmembrane glycoprotein, broadly expressed among hematopoietic cells. Multiple isoforms of CD45R are distributed throughout the immune system according to cell type. These isoforms arise because of alternative splicing of exons 4, 5, and 6. The corresponding protein domains are characterized by the binding of monoclonal antibodies specific for CD45RA (exon 4), CD45RB (exon 5), CD45RC (exon 6) and CD45RO (exons 4 to 6 spliced out). The variation in these isoforms is localized to the extracellular domain of CD45R, while the intracellular domain is conserved. Renalase is a 342 amino acid FAD-dependent amine oxidase that is highly expressed in kidney and is expressed at a lower level in heart, skeletal muscle and small intestine. Renalase is secreted in the blood by the kidney and it is thought to regulate cardiac function and systemic blood pressure. It is also suggested that Renalase functions as a hormone that metabolizes circulating catecholamines, which have an active role in the sympathetic and parasympathetic nervous systems. Individuals with chronic kidney disease and end-stage renal disease have markedly reduced levels of plasma Renalase than healthy individuals. Infusion of Renalase in animal models causes decrease in heart rate, cardiac contractility and blood pressure. Recognizes a polypeptide which is identified as insulin, a 51-amino acid polypeptide composed of A and B chains connected through the C-peptide. Proinsulin, which has very little biological activity, is cleaved by proteases within its cell of origin into the insulin molecule and the C-terminal basic residue. Insulin enhances membrane transport of glucose, amino acids, and certain ions. It also promotes glycogen storage, formation of triglycerides, and synthesis of proteins and nucleic acids. This Ab reacts with the rod domain of human cytokeratin 19 (CK19), a polypeptide of 40 kDa. CK19 is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. Spectrin is an actin binding protein that is a major component of the plasma membrane skeleton. Spectrins function as membrane organizers and stabilizers by forming dimers, tetramers and higher polymers. Vertebrate spectrins have two alpha-subunits (alpha-I/alpha-II), four beta-subunits (beta-I-beta-IV) and a beta-H subunit creating diversity and specialization of function. Spectrin alpha and spectrin beta are present in erythrocytes, whereas spectrin alpha II (also designated fodrin alpha) and spectrin beta I (also designated fodrin beta) are present in other somatic cells. The spectrin tetramers in erythrocytes act as barriers to lateral diffusion, but spectrin dimers seem to lack this function. This MAb recognizes a 21 kDa protein, identified as the p21WAF1 tumor suppressor protein. This MAb is highly specific to p21 and shows no cross-reaction with other closely related mitotic inhibitors. p21WAF1 is a specific inhibitor of cdk's and a tumor suppressor involved in the pathogenesis of a variety of malignancies. The expression of this gene acts as an inhibitor of the cell cycle during G1 phase and is tightly controlled by the tumor suppressor protein p53. Its expression is induced by the wild type, but not mutant, p53 suppressor protein. Normal cells generally display a rather intense nuclear p21 expression. CD9 is a type IV transmembrane glycoprotein with four transmembrane domains. CD9 on pre-B cells may play a role in cell-cell adhesion. In addition, CD9 may play a role in signal transduction mediated by interaction with low molecular weight GTP binding proteins. CD9 is expressed on early B cells, eosinophils, basophils and activated T cells and is a major component of the platelet cell surface. Alpha actinins belong to the spectrin gene superfamily which represents a diverse group of cytoskeletal proteins, including the alpha and beta spectrins and dystrophins. Recognizes a protein of 110 kDa, identified as Excision Repair Cross Complementing 1 (ERCC1). It is a mammalian nucleotide excision repair (NER) enzyme involved in repair of damaged DNA. ERCC1 is a homologous to RAD10 in Saccharomyces cerevisiae, which is required in mitotic intrachromosomal recombination and repair. Cytokeratin 13 (KRT13) is the major acidic keratin, which together with KRT4, its basic partner, is expressed in the suprabasal layers of non-cornified stratified epithelia including tongue mucosa, esophagus, anal canal epithelium, tracheal epithelium, uterine cervix, and urothelium. Defects in the KRT13 gene are a cause of white sponge nevus of cannon (WSN), a rare autosomal dominant disorder, which predominantly affects non-cornified stratified squamous epithelia and is characterized by the presence of soft, white and spongy plaques in the oral mucosa.
