Biotium Prohibitin (Mitochondrial Marker)(PHB/3226), CF740 conjugate, 0.1mg/mL
Recognizes a protein of 30 kDa which is identified as Prohibitin, an evolutionarily conserved protein with homologues found in yeast to man. It is located in the inner membrane of mitochondria. Although prohibitin mRNA and protein expression occurs throughout the cell cycle, maximum levels are detected during the G1/S phase transition and minimum levels are seen in S phase and the G2/mitosis boundary. Prohibitin is located exclusively in the mitochondria with the highest concentration on the inner membrane. Prohibitin is an ideal mitochondrial marker. It shows antiproliferative activity and has been proposed to play a role in normal cell cycle regulation, replicative senescence, cellular immortalization, and tumor suppression. Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. CF Dyes offer exceptional brightness and photostability. Note: Conjugates of blue fluorescent dyes like CF405S and CF405M are not recommended fo

Biotium Cyclin B1 (G2- & M-phase Cyclin)(CCNB1/2978R), CF568 conjugate, 0.1mg/mL
Recognizes a protein of 55-62 kDa, identified as cyclin B1. In mammals, cyclin B associates with inactive p34cdc2, which facilitates phosphorylation of p34cdc2 at aa 14Thr and 15Tyr. This maintains the inactive state until the end of G2-phase. The inactive cyclin B-p34cdc2 complex continues to accumulate in the cytoplasm until the completion of DNA synthesis, when Cdc25, a specific protein phosphatase, dephosphorylates aa 14Thr and 15Tyr of p34cdc2 rendering the complex active at the G2/M boundary. This mitotic kinase complex remains active until the metaphase/anaphase transition when cyclin B is degraded. This degradation process is ubiquitin-dependent and is necessary for the cell to exit mitosis. So, cyclin B-p34cdc2 plays a critical role in G2 to M transition. Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. CF Dyes offer exceptional brightness and photostability. Biotium Cytochrome C (Mitochondrial Marker)(7H8.2C12 + CYCS/1010), 1mg/mL
Cytochrome C is a well-characterized mobile electron transport protein that is essential to energy conversion in all aerobic organisms. In mammalian cells, this highly conserved protein is normally localized to the mitochondrial inter-membrane space. More recent studies have identified cytosolic cytochrome c as a factor necessary for activation of apoptosis. During apoptosis, cytochrome c is trans-located from the mitochondrial membrane to the cytosol, where it is required for activation of caspase-3 (CPP32). Overexpression of Bcl-2 has been shown to prevent the translocation of cytochrome c, thereby blocking the apoptotic process. Overexpression of Bax has been shown to induce the release of cytochrome c and to induce cell death. The release of cytochrome c from the mitochondria is thought to trigger an apoptotic cascade, whereby Apaf-1 binds to Apaf-3 (caspase-9) in a cytochrome c-dependent manner, leading to caspase-9 cleavage of caspase-3. This MAb recognizes total cytoch

Biotium CD99 / MIC2 (Ewing's Sarcoma Marker), CF568 conjugate, 0.1mg/mL
Recognizes a sialoglycoprotein of 27-32 kDa, identified as CD99, or MIC2 gene product, or E2 antigen. MIC2 gene is located in the pseudo-autosomal region of the human X and Y chromosome. MIC2 gene encodes two distinct proteins, which are produced by alternative splicing of the CD99 gene transcript and are identified as bands of 30 and 32 kDa (p30/32). Although its function is not fully understood, CD99 is implicated in various cellular processes including homotypic aggregation of T cells, upregulation of T cell receptor and MHS molecules, apoptosis of immature thymocytes and leukocyte diapedesis. CD99 is expressed on the cell membrane of some lymphocytes, cortical thymocytes, and granulosa cells of the ovary. Most pancreatic islet cells, Sertoli cells of the testis, and some endothelial cells express this antigen. Mature granulocytes express very little or no CD99. MIC2 is strongly expressed on Ewing's sarcoma cells and primitive peripheral neuroectodermal tumors. Primary ant

Biotium Erythropoietin (EPO) (EPO/1367), CF740 conjugate, 0.1mg/mL
This antibody recognizes a protein of about 37 kDa, which is identified as Erythropoietin (EPO). Erythropoietin is a secreted, glycosylated cytokine hormone composed of four alpha helical bundles. It is the primary factor responsible for regulating erythropoiesis during steady-state conditions and in response to blood loss and hemorrhage in the adult organism. Erythropoietin is synthesized by the kidney and stimulates the proliferation and maturation of bone marrow erythroid precursor cells. The protein is found in the plasma and regulates red cell production by promoting erythroid differentiation and initiating hemoglobin synthesis.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. CF Dyes offer exceptional brightness and photostability. Note: Conjugates of blue fluorescent dyes like CF405S and CF405M are not recommended for detecting low abundance targets, because blue dyes have lower fluorescence and can give higher

Biotium ROR-gamma / RORC (RAR-related Orphan Receptor C)(RORC/2942), CF568 conjugate, 0.1mg/mL
This MAb recognizes a protein of 63 kDa, identified as ROR-C. Its epitope maps in between aa1-50. The nuclear orphan receptors ROR and ROR are members of the nuclear hormone receptor superfamily. Members of this family act by directly associating with DNA sequences known as hormone response elements (HREs) and typically bind DNA as either homo- or heterodimers. RORalpha and RORgamma are unique in that they bind DNA as monomers. RORalpha has multiple isoforms that share common DNA and putative ligand-binding domains, but differ in their amino terminal domains, which are generated by alternative RNA processing. RORgamma comprises a 560 amino acid protein that shares 50% amino acid identity with RORalpha and is most highly expressed in skeletal muscle. Although these proteins are considered orphan receptors, due to a lack of defined ligands, experimental evidence has shown that melatonin may be the natural ligand for these nuclear receptors. Primary antibodies are available puri

Biotium HPV-16 E6 + HPV-18 E6 (Human Papilloma Virus 16/18)(HPV16/1295 + HPV18/1297), CF488A conjugate, 0.1mg/mL
Human papilloma viruses (HPVs) can be classified as either high risk or low risk according to their association with cancer. HPV16 and HPV18 are the most common of the high risk group while HPV6 and HPV11 are among the low risk types. Approximately 90% of cervical cancers contain HPV DNA of the high risk types. Mutational analysis has shown that the E6 and E7 genes of the high risk HPVs are necessary and sufficient for HPV transforming function. The specific interactions of the E6 and E7 proteins with p53 and pRB, respectively, correlate with HPV high and low risk classifications. The high risk HPV E7 proteins bind to pRB with a higher affinity than do the low risk HPV proteins, and only the high risk HPV E6 proteins form detectable complexes with p53 in vitro. Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. CF Dyes offer exceptional brightness and photostability. Note: Conjugates of blue fluorescent dyes like CF405S

Biotium Chromogranin A / CHGA (Neuroendocrine Marker), 0.2mg/mL
Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Most pituitary adenomas and prolactinomas readily express chromogranin.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. Biotium ARF1 (Golgi Apparatus Marker)(1A9/5), Biotin conjugate, 0.1mg/mL
The ADP-ribosylation factor (ARF) family comprises a group of structurally and functionally conserved proteins, which are members of the Ras superfamily of regulatory GTP-binding proteins. The ARF family is divided functionally into the ARF and the ARF-like proteins. ARF's share more than 60% sequence identity, appear to be ubiquitous in eukaryotes, and are highly conserved evolutionarily. ARF is involved in intracellular protein traffic to and within the Golgi complex. ARF has a number of disparate activities including maintenance of organelle integrity, assembly of coat proteins, as a co-factor for cholera toxin and as an activator of phospholipase D. Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. CF Dyes offer exceptional brightness and photostability. Biotium N-Cadherin / Cadherin-2 / CD325 (NCAD) (13A9), CF488A conjugate, 0.1mg/mL
This antibody recognizes a protein of ~140 kDa, identified as N-Cadherin (NCAD), also known as CD325. NCAD is a member of the Cadherin superfamily, and consists of five extracellular repeats, a transmembrane domain and a cytoplasmic domain. CD325 deficient mice die at day 10 of gestation and embryos display major heart defects and malformed neural tubes and somites. Consistent with this, CD325 has been implicated in several aspects of cardiac development including the precardiac mesoderm, establishment of left-right symmetry and cardiac looping morphogenesis. Furthermore, CD325 is normally involved in inducing cell cycle arrest and its expression is frequently deregulated in cancer cells. Studies have linked N-cadherin to cancer metastasis by showing the aggressive tumor cells had preferentially turned on N-cadherin as opposed to E- or P-cadherin.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. Biotium DOG-1 / TMEM16A (Gastrointestinal Stromal Tumor Marker) (DG1/1486), CF740 conjugate, 0.1mg/mL
Expression of DOG-1 protein is elevated in the gastrointestinal stromal tumors (GIST s), c-kit signaling-driven mesenchymal tumors of the GI tract. DOG-1 is rarely expressed in other soft tissue tumors, which, due to appearance, may be difficult to diagnose. Immunoreactivity for DOG-1 has been reported in 97.8 percent of scorable GIST s, including all c-kit negative GIST s. Overexpression of DOG-1 has been suggested to aid in the identification of GISTs, including Platelet-Derived Growth Factor Receptor Alpha mutants that fail to express c-kit antigen. The overall sensitivity of DOG1 and c-kit in GIST s is nearly identical: 94.4% vs. 94.7%.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. Biotium MUC1 / CA15-3 / EMA / CD227 (Epithelial Marker)(MUC1/2818R), CF488A conjugate, 0.1mg/mL
This MAb reacts with MUC1. The dominant epitope of this MAb has not yet been determined. MUC1 is a large cell surface mucin glycoprotein expressed by most glandular and ductal epithelial cells and some hematopoietic cell lineages. It is expressed on most secretory epithelium, including mammary gland and some hematopoietic cells. It is expressed abundantly in lactating mammary glands and over expressed abundantly in 90% breast carcinomas and metastases. Transgenic MUC1 has been shown to associate with all four c-erbB receptors and localize with c-erbB1 (EGFR) in lactating glands. The MUC1 gene contains seven exons and produces several different alternatively spliced variants. The major expressed form of MUC1 uses all seven exons and is a type 1 transmembrane protein with a large extracellular tandem repeat domain. The tandem repeat domain is highly O glycosylated and alterations in glycosylation have been shown in epithelial cancer cells. Antibody to EMA is useful as a pan-epi

Biotium Smooth Muscle Actin(1A4), CF740 conjugate, 0.1mg/mL
Actin is a major component of the cytoskeleton and is present in most cell types. This MAb is highly specific to actin from smooth muscles. Its epitope lies in the first four N-terminal amino acids. This MAb does not stain cardiac or skeletal muscle; however, it does stain myofibroblasts and myoepithelial cells. This antibody could be used together with anti-muscle specific actin and myogenin in making a diagnosis of smooth muscle and skeletal muscle tumors. In most cases of rhabdomyosarcoma, this antibody yields negative results whereas anti-muscle specific actin and myogenin are positive. Leiomyosarcomas are positive only with anti-muscle specific actin and anti-smooth muscle actin and are negative with anti-myogenin.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels.
