Recognizes a protein of 30-33 kDa, which is identified as CD20. It is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre-B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells. This MAb can be used for immunophenotyping of leukemia and malignant cells, B lymphocyte detection in peripheral blood and B cell localization in tissues. It reacts with the majority of B-cells present in peripheral blood and lymphoid tissues and their derived lymphomas. In lymphoid tissue, germinal center blasts and B-immunoblasts are particularly reactive. It is a reliable antibody for ascribing a B-cell phenotype in known lymphoid tissues. The protein encoded by this gene is a member of the transmembrane 4 superfamily, also known as the tetraspanin family. Most of these members are cell-surface proteins that are characterized by the presence of four hydrophobic domains. The proteins mediate signal transduction events that play a role in the regulation of cell development, activation, growth and motility. This encoded protein is found in the asymmetrical unit membrane (AUM) where it can form a complex with other transmembrane 4 superfamily proteins. It may play a role in normal bladder epithelial physiology, possibly in regulating membrane permeability of superficial umbrella cells or in stabilizing the apical membrane through AUM/cytoskeletal interactions. Spectrin is an actin binding protein that is a major component of the plasma membrane skeleton. Spectrins function as membrane organizers and stabilizers by forming dimers, tetramers and higher polymers. Vertebrate spectrins have two alpha-subunits (alpha-I/alpha-II) four beta-subunits (beta-I-beta-IV) and a beta-H subunit creating diversity and specialization of function. Spectrin III is highly expressed in brain, kidney, pancreas and liver, and at lower levels in lung and placenta. This MAb recognizes a protein of 170 kDa, identified as EGFR. EGFR is type I receptor tyrosine kinase with sequence homology to erbB-1, -2, -3 -4 or HER-1, -2, -3 -4. It binds to Epidermal Growth Factor (EGF), Transforming Growth Factor-a (TGF-a), Heparin-binding EGF (HB-EGF), amphiregulin, betacellulin and epiregulin. Recognizes Nucleolin (NCL), the major nucleolar phosphoprotein of growing eukaryotic cells. NCL is located mainly in dense fibrillar regions of the nucleolus. It is found associated with intranucleolar chromatin and pre-ribosomal particles. Human NCL gene consists of 14 exons with 13 introns and spans approximately 11 kb. It has a predicted molecular weight of ~76 kDa with an apparent MW of 100-110 kDa, attributed to phosphorylation of its N-terminal domain. It induces chromatin decondensation by binding to histone H1. Decorin is a small leucine-rich proteoglycan (SLRP) family member that consists of a glycosaminoglycan chain-containing core protein. The core protein contains ten leucine rich repeats that contain sites for glycosylation, flanked by disulfide bond stabilizing loops. Decorin binds to Collagen Type I, II and IV in vivo and promotes the formation of fibers with variations in stability and solubility. The Decorin core protein binds to growth factors, intercellular matrix molecules, such as Fibronectin and Thrombospondin, and to the Decorin endocytosis receptor. Decorin binds to and inhibits TGF and is a direct or indirect negative modulator of TGF synthesis. Inhibition of Decorin core protein gene expression by the combination of IFN- and TNF may contribute to cartilage destruction that is characteristic of inflammatory joint diseases.
