Creatine kinases (CK) are a large family of isoenzymes that regulate levels of ATP in subcellular compartments, where they provide ATP at sites of fluctuating energy demand by the transfer of phosphates between creatine and adenine nucleotides. CKs provide the energy of phosphate hydrolysis necessary to drive the normal function of many cellular systems. In cells, the cytosolic CK enzymes consist of two subunits, which can be either B (brain type) or M (muscle type). There are three different isoenzymes: CKMM, CKBB and CKMB. This MAb recognizes the CKBB isoenzyme and does not react with the B subunit in CKMB. Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. This MAb recognizes a 21 kDa protein, identified as the p21WAF1 tumor suppressor protein. This MAb is highly specific to p21 and shows no cross-reaction with other closely related mitotic inhibitors. p21WAF1 is a specific inhibitor of cdk's and a tumor suppressor involved in the pathogenesis of a variety of malignancies. The expression of this gene acts as an inhibitor of the cell cycle during G1 phase and is tightly controlled by the tumor suppressor protein p53. Its expression is induced by the wild type, but not mutant, p53 suppressor protein. Normal cells generally display a rather intense nuclear p21 expression. Loss of p21 expression has been reported in many carcinomas (gastric carcinoma, non-small cell lung carcinoma, thyroid carcinoma). Prostaglandins are a diverse group of autocrine and paracrine hormones that mediate many cellular and physiologic processes. Prostaglandin H2 (PGH2) is an intermediate molecule in formation of the prostaglandins. Cyclooxygenase-1 (Cox-1) and cyclooxygenase-2 (Cox-2) are prostaglandin synthases that catalyze the formation of PGH2 from arachidonic acid (AA). Cox-1 and Cox-2 are isozymes of prostaglandin-endoperoxidase synthase (PTGS). Cox-1 is constitutively expressed in most tissues and is thought to serve in general housekeeping functions. Cox-2 is efficiently induced in migratory cells responding to pro-inflammatory stimuli and is considered to be an important mediator of inflammation. CLP36, also known as PDLIM1, CLIM1 or Elfin, is a 329 amino acid cytoplasmic protein that associates with actin stress fibers at the cytoskeleton. Expressed at high levels in skeletal muscle and heart and at lower levels in colon, small intestine, spleen, lung, placenta, kidney, liver, thymus and pancreas, CLP36 functions as a cytoskeletal protein that is thought to act as an adaptor, bringing target proteins to the cytoskeleton. Specifically, CLP36 interacts with Clik1 (-actinin-1, thereby facilitating the association of Clik1 with actin stress fibers. CLP36 contains one PDZ domain and one LIM zinc-binding domain through which it conveys its protein-protein binding capabilities. Human CLP36 shares 88% sequence similarity with its rat counterpart, suggesting a conserved function between species. Succinate dehydrogenase (SDH) is Complex II in the mitochondria, vital for mitochondrial electron transport, as well as Krebs cycle function. Four subunits comprise the SDH protein complex: a flavochrome subunit (SDHA), an iron-sulfur protein (SDHB) and two membrane-bound subunits (SDHC and SDHD) anchored to the inner mitochondrial membrane. The SDH complex functions as a tumor suppressor. Loss of any subunit proteins lead to destabilization of the complex and tumor formation. Antibody to SDHB is helpful in the identification of phaeochromocytomas, paragangliomas and GIST. Biotium Tyrosinase (Melanoma Marker)(rOCA1/812), CF488A conjugate, 0.1mg/mL
Recognizes a cluster of proteins between 70-80 kDa, identified as tyrosinase. Occasionally a minor band at 55 kDa is also detected. This MAb shows no cross-reaction with MAGE-1 and tyrosinase-related protein 1, TRP-1/gp75. Tyrosinase is a copper-containing metalloglycoprotein that catalyzes several steps in the melanin pigment biosynthetic pathway; the hydroxylation of tyrosine to L-3, 4-dihydroxy-phenylalanine (dopa), and the subsequent oxidation of dopa to dopaquinone. Mutations of the tyrosinase gene occur in various forms of albinism. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. Staining of melanomas with this MAb shows tyrosinase in melanotic as well as amelanotic variants. This MAb is a useful marker for melanocytes and melanomas.
