This MAb recognizes a protein of 170 kDa, identified as EGFR. EGFR is type I receptor tyrosine kinase with sequence homology to erbB-1, -2, -3 -4 or HER-1, -2, -3 -4. It binds to Epidermal Growth Factor (EGF), Transforming Growth Factor-a (TGF-a), Heparin-binding EGF (HB-EGF), amphiregulin, betacellulin and epiregulin. EGFR is overexpressed in tumors of breast, brain, bladder, lung, gastric, head & neck, esophagus, cervix, vulva, ovary, and endometrium. SOX2 is required for stem cell maintenance in the central nervous system, and it also regulates gene expression in the stomach. Complement C1q B Chain (C1qb) is a subunit of C1q. C1q is composed of 6 polypeptide chains each of C1qa, C1qb, and C1qc. C1q is a component of the classical complement pathway, and thereby plays an important role in the immune response. Each C1q subunit contains an N-terminal collagen-like region and a C-terminal C1q globular domain. The presence of receptors for C1q on effector cells modulates its activity, which may be antibody-dependent or independent. Calbindin 1, also known as calbindin D28K, is a member of a family of calcium binding proteins. This protein family also includes calmodulin, calbindin 2, S-100, and troponin C. Calbindin 1 is a 261-amino acid protein with 6 EF-hand domains, 4 of which are active calcium-binding domains. Expressed in brain, ovary, uterus, testis, pancreas, liver, kidney and intestine, calbindin 1 acts as a calcium-buffering agent and alters the activity of the plasma membrane ATPase. In neuronal cells, calbindin 1 modulates calcium channel activity, calcium transients and intrinsic neuronal firing activity. GP2 (glycoprotein 2), also known as ZAP75, is a 537 amino acid secreted protein. It is an integral membrane protein that is secreted from intracellular zymogen granules and associates with the plasma membrane via glycosylphosphatidylinositol (GPI) linkage. GP2 is cleaved and then released into the pancreatic duct along with exocrine secretions. GP2 binds pathogens such as enterobacteria, thereby playing an important role in the innate immune response. The C-terminus of this protein is related to the C-terminus of the protein encoded by the neighboring gene, uromodulin (UMOD). This antibody recognizes a protein of 42 kDa, which is identified as Alpha-methylacyl-CoA Racemase (AMACR), also known as p504S. It is an enzyme that is involved in bile acid biosynthesis and -oxidation of branched-chain fatty acids. AMACR is essential in lipid metabolism. It is expressed in cells of premalignant high-grade prostatic intraepithelial neoplasia (HGPIN) and prostate adenocarcinoma. MAb r2H11 reacts with a partially defined epitope of human thyroglobulin. This epitope is different form the epitope recognized by MAb 6E1. Thyroglobulin is a 660 kDa dimeric pre-protein with mutiple glycosylation sites. It is produced by and processed within the thyroid gland to produce the hormone thyroxine and triiodothyronine. Prior to forming dimers, thyroglobulin monomers undergo conformational maturation in the endoplasmic reticulation. The vast majority of follicular carcinomas of the thyroid will give positive immunoreactivity for anti-thyroglobulin even though sometimes only focally. Poorly differentiated carcinomas of the thyroid are frequently anti-thyroglobulin negative. Adenocarcinomas of other-than-thyroid origin do not react with this antibody. Recognizes a protein of 30-33 kDa, which is identified as CD20. It is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre-B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells.
