Enzo Life Sciences Nupherin™ Transfection Reagent (3 mg)
Nupherin™ dramatically enhances the performance of commercially available cationic lipids to allow high-efficiency lipofection (10-40%) of neuronal, ganglion and other difficult to transfect cells. Efficient lipofection is limited by the ability of transfected DNA to enter the nucleus – a particular obstacle in nondividing or slow dividing cells like neurons. Nupherin, a peptide based reagent that combines a non-classical nuclear localization sequence (M9) with a nucleotide binding domain, helps to transport DNA into the transfected cell's nucleus. Nupherin has been demonstrated to provide high efficiency lipofection of primary rat hippocampal neurons, chicken embryonic retinal ganglian cells, rat pheochromocytoma (PC12) cells and other recalcitrant cells. Fully compatible with cells grown on glass coverslips, in plastic plates, or in tissue-culture flasks. Formulation: Liquid. Sterile solution. Long Term Storage: -80°C.

The Expression Media Optimization Kit™, enables researchers to determine the best media formulation that yields the highest amount of target protein. Since the production of a given recombinant protein can vary significantly depending on the type of medium employed, the EMSK™ provides sterile liquid aliquots of fifteen expression broths, including eleven proprietary formulations, as well as Animal Product Free(APF) versions of Miller and Lennox. ReproRNA™-OKSGM is a single-stranded RNA replicon vector that contains five reprogramming factors: OCT4, KLF-4, SOX2, GLIS1, and c-MYC, as well as a puromycin-resistance gene (Figure 1). This RNA vector reprograms somatic cells, such as fibroblasts, into induced pluripotent stem (iPS) cells with high efficiency and only requires a single transfection step (Figure 2). When used together with ReproTeSR™ reprogramming medium, the generation of iPS cell colonies can be achieved under feeder-free conditions with superior colony morphology and similar reprogramming efficiency to feeder-based systems (Figures 3 - 4). ReproRNA™-derived iPS cell colonies also express markers of undifferentiated cells and retain a normal karyotype (Figures 5-6). Subsequently, iPS cells generated with ReproRNA™-OKSGM can be maintained in TeSR™ maintenance media and further differentiated into cells of all three germ layers (Figure 7).
