pADL™-100 is a phagemid vector designed for high levels of display on the filamentous bacteriophage M13 or equivalent. This vector contains a PelB leader sequence followed by a full-length copy of the gene III sequence and a double-SfiI cloning site to introduce foreign polypeptides on the N-terminal side of the gene III protein. Expression of the fusion is under the tight control of a lac promoter. Ad infection is initiated by recognition of the native Ad5 receptor, coxsackievirus-adenovirus receptor (CAR), on target cells by the carboxy-terminal portion (i.e., knob) of the fiber protein. Therefore the infection efficiency of adenovirus is very low for cells with low or no CAR receptors. Arg-Gly-Asp (RGD) motif was introduced in the surface-exposed loops of adenovirus fiber knob, this enables the adenovirus to bypass CAR and mediate cell entry via RGD binding integrins. So the RDG modified adenovirus can be used for "retargeting" cells that regular adenovirus doesn't work well. The infection effeciency of RDG modified adenovirus can be tens or hundreds of times higher than standard Ad5 in some cells, including human or mouse macrophages, T cells, synoviocytes, islet grafts, adipocytes, MEF, etc.This is a RGD modified replication-deficient adenovirus expressing eGFP under a CMV promoter.

Heterotrimeric G protein-mediated signal transduction is a dynamically regulated process with the intensity of signal decreasing over time despite the continued presence of the agonist. This phenomenon, referred to as agonist-mediated desensitization, involves phosphorylation of the receptor by two classes of enzymes. The first is the second messenger regulated kinases such as cAMP dependent protein kinase A and protein kinase C. The second is the G protein-coupled receptor kinases (GRKs). Six members of the GRK family have been identified to date. These include rhodopsin kinase (GRK 1), two forms of beta-adrenergic receptor kinase (GRK 2 or beta-ARK 1 and GRK 3 or beta-ARK 2), IT-11 (GRK 4), GRK 5 and GRK 6. Phosphorylation of receptors by GRKs appears to be strictly dependent on the receptor being in its agonist-activated state.
