Genetic and biochemical studies have led to the identification of the Stat3-Interacting Protein STATIP1. The preferential association of STATIP1 with inactive (i.e., unphosphorylated) Stat3 suggests that it may contribute to the regulation of Stat3 activation. Overexpression of the Stat3-binding domain of STATIP1 blocks Stat3 activation, nuclear translocation, and Stat3-dependent induction of a reporter gene.

This gene encodes a member of the BCL-2 protein family. The proteins of this family form hetero- or homodimers and act as anti- and pro-apoptotic regulators that are involved in a wide variety of cellular activities such as embryonic development, homeostasis and tumorigenesis. The protein encoded by this gene is able to reduce the release of pro-apoptotic cytochrome c from mitochondria and block caspase activation. This gene is a direct transcription target of NF-kappa B in response to inflammatory mediators, and has been shown to be up-regulated by different extracellular signals, such as granulocyte-macrophage colony-stimulating factor (GM-CSF), CD40, phorbol ester and inflammatory cytokine TNF and IL-1, which suggests a cytoprotective function that is essential for lymphocyte activation as well as cell survival.

The endothelins are family of cysteine-rich peptides consisting of 21-amino acid and containing two intramolecular disulfide bridges that exert their vasoactive and mitogenic actions through G protein-coupled receptors. At the present, the existence of three active isoforms of ET (-1, -2, and -3), and two distinct receptors, ETA and ETB, is well documented (17). The active form of ET-1 arises from biologically inactive big-ET-1 through the action of a specific protease that removes 18 amino acids from its carboxyl terminal. This membrane-bound metallopeptidase, characterized by its sensitivity to phosphoramidon, is known as endothelin-converting enzyme-1 (ECE-1) . A growing body of evidence indicate that ET-1 is a major player in numerous disease states including congestive heart failure, pulmonary fibrosis, obesity and diabetes, as well as a key mediator of tissue repair and normal wound healing.

Aurora related kinase-1 (ARK-1, STK15, STK6, Aurora2) and -2 (ARK-2, STK12, Aurora1) are centrosome-associated serine/threonine kinases that regulate centrosome separation, bipolar spindle assembly and chromosome segregation during mitosis. First characterized in human colorectal cancers, ARK-1 and -2 are closely related to Drosophila aurora kinases. ARK-1 protein has elevated expression in colon carcinoma lines (HT-29, SNU-C2B, COLO 205, SW480, 837 and 948) and accumulates during metaphase in HeLa cells. ARK-1 and -2 have unique genetic loci, suggesting that these two kinases, with oncogenic potential, play different roles in cell cycle progression.

pCSG-IBAwt2 is a large expression vector with universal features for transient expression as well as for generation of stable mammalian cell lines. The vector carries an ampicillin resistance cassette for selection of transformed E. coli cells and ColE1 origin for a high plasmid copy number. Extrachromosomal replication in mammalian cells could occur either by origin of replication from Epstein-Barr Virus (oriP) or by SV40 ori. For the former the vector provides the EBNA-1 gene and for the latter the cell line has to be latently infected with SV40 or express the SV40 large T antigen (e.g. HEK293T, COS-1, COS-7). Stable cell lines can be selected by the neomycin resistance gene (NeoR). In addition, the human cytomegalovirus (CMV) immediate-early promoter enables a high-level expression. During translocation from the cytosol the signal peptide is removed from the protein by endogenous proteases.
