Cas9 is the nuclease guided by the crRNA and tracrRNA (or trans-activating crRNA) to cleave specific DNA sequences. A guide RNA (gRNA) can be designed to include a hairpin that mimics the tracrRNA-crRNA complex. Binding specificity is based on the gRNA and a three nucleotide NGG sequence called the protospacer adjacent motif (PAM) sequence. With its ease in designing guide sequences to target specific genomic loci, the CRISPR/Cas system is a much simpler, faster, and robust alternative to TALEN and Zinc finger nuclease platforms. For site-specific genome editing, the CRISPR/Cas9 system requires the Cas 9 nuclease and the gRNA. This adenovirus expresses a codon-optimized Cas9 nuclease, along with a mCherry reporter. It can be used along with guided RNA for genome-editing.This adenovirus expresses a FLAG tagged Cas9 with a mCherry reporter.

The pASG-IBA143 vector is designed for expression of recombinant proteins with an N-terminal His-tag and C-terminal Twin-Strep-tag in E. coli. The vector carries an ampicillin resistance cassette for selection of transformed E. coli cells, F1 and ColE1 origin for a high plasmid copy number, and the inducible tetracycline promoter/operator for the regulated expression of proteins. Expression of the recombinant protein is induced by addition of anhydrotetracycline, which is a derivative of tetracycline that does not exhibit antibiotic activity. In addition to the direct cloning of the gene of interest into pASG-IBA vectors with Esp3I, another option via a so-called Entry Vector is possible.

Syk p72 and ZAP-70 resemble the Src family protein tyrosine kinases in that they have a C-terminal catalytic domain, but differ in that they are characterized by two SH2 domains but no SH3 domains. Syk p72 kinase fused to the transmembrane and extracellular domains of CD7 and CD16, respectively, can induce complete T cell activation, implying the involvement of the Syk/ZAP family proteins in T cell activation. In contrast, the ZAP-70 kinase is insufficient for complete T cell activation unless it is coaggregated with a Fyn p59-containing chimera, suggesting that regulation of ZAP-70 activity may require a functional interaction with Src family kinases.
