The protein encoded by this gene belongs to the PI3/PI4-kinase family. This protein is an important cell cycle checkpoint kinase that phosphorylates; thus, it functions as a regulator of a wide variety of downstream proteins, including tumor suppressor proteins p53 and BRCA1, checkpoint kinase CHK2, checkpoint proteins RAD17 and RAD9, and DNA repair protein NBS1. This protein and the closely related kinase ATR are thought to be master controllers of cell cycle checkpoint signaling pathways that are required for cell response to DNA damage and for genome stability. Mutations in this gene are associated with ataxia telangiectasia, an autosomal recessive disorder.

The myocyte enhancing factor-2 (MEF-2) family of transcription factors includes four splice variants A, B, C and D which encode muscle-specific DNA binding proteins that recognize an A/T rich sequence [CTA (A/T)4 TAG] localized in the control regions of numerous muscle-specific genes.

The cyclic nucleotide phosphodiesterases (PDEs) have an important role to play in the regulation of cyclic nucleotide levels, as PDE activity hydrolyses camp and cGMP [1-5]. The PDEs are a large superfamily of enzymes consisting of eleven discovered isozyme families (PDE1-11) that differ in respect to substrate specificity, subcellular distribution and regulatory properties [2, 3, 6-9]. Of these eleven families, cGMP-activated PDE (PDE3), cAMP-specific PDE (PDE4), and the more recently discovered PDE7 and PDE8 isozymes mainly hydrolyse cAMP. The PDE5, PDE6 and PDE9 isozymes prefer cGMP as a substrate. Calcium/calmodulin-stimulated PDE (PDE1), cGMP-stimulated PDE (PDE2), PDE10 and PDE11 are dual-substrate PDEs that hydrolyse both cAMP and cGMP.
