IBA LifeSciences StrGate Acptr Vctr pESG-IBAwt1
The pESG-IBAwt1 vector is designed for high-level, stable, and non-replicative transient expression of target proteins without affinity tag in mammalian cells. The vector carries an ampicillin resistance cassette for selection of transformed E. coli cells as well as the F1 and ColE1 origin for a high plasmid copy number. In addition, it carries the human cytomegalovirus (CMV) immediate-early promoter for high-level expression in a wide range of mammalian cells, the neomycin resistance gene for selection of stable cell lines, and the SV40 ori for episomal replication in cell lines that are latently infected with SV40 or that express the SV40 large T antigen (e.g. HEK293T, COS-1, COS-7). In addition to the direct cloning of the gene of interest into pESG-IBA vectors with Esp3I, another option via a so-called Entry Vector is possible.

The Exontrap vector pET01 has an intrinsic splicing function that enables selective cloning of exon sequences from large genomic eukaryotic DNA fragments. This new route for the identification of eukaryotic genes does not involve an initial isolation of cellular mRNA, allowing genes that are not transcribed during certain life cycle stages to be identified. This greatly facilitates exon/intron mapping. The Exontrap function is based on a shuttle vector containing prokaryotic and eukaryotic genetic elements for replication in both bacteria and cell cultures. The vector contains a 5´ and 3´ exon separated by a 600 bp intron sequence that has a polylinker for cloning.

Protein tags are peptide sequences genetically grafted onto a recombinant protein. Often these tags are removable by chemical agents or by enzymatic means, such as proteolysis or intein splicing. Tags are attached to proteins for various purposes.Epitope tags are short peptide sequences which are chosen because high-affinity antibodies can be reliably produced in many different species. These are usually derived from viral genes, which explain their high immunoreactivity. Epitope tags include V5-tag, Myc-tag, HA-tag and NE-tag. These tags are particularly useful for western blotting, immunofluorescence and immunoprecipitation experiments, although they also find use in antibody purification.
