An HA tag was fused in frame to the N termini of the mouse Akt1 coding sequences.Akt1/PKB is activated in response to many extracellular signals and play key roles in regulating diverse cellular processes, including cell proliferation, differentiation, apoptosis, angiogenesis and glycogen synthesis etc. Akt1 is activated by the phosphorylation of the T loop of their kinase domain by PDK1 and by phosphorylation of a residue located C-terminal to the kinase domain in the hydrophobic motif. The Akt family of kinases includes Akt1, Akt2 and Akt3.

A mutated form of estrogen ligand-binding domain (ERT2) that binds to synthetic antagonists (such as tamoxifen or its derivative 4-hydroxy-tamoxifen) but not to circulating estrogens was fused to the Cre recombinase (Cre) to create the CreERT2, which requires the presence of tamoxifen for Cre recombinase activity. Therefore it makes possible to induce the recombination in cells with tamoxifen.This AAV-1 virus expresses CreERT2 under a CMV promoter.

Cre recombinase is used as a tool to genetically modify genes, such as to delete a segment of DNA flanked by LoxP sites in cells or experimental animals.By applying the mammalian codon usage to Cre recombinase, expression of Cre is improved in the mammalian cells. This improved Cre (iCre) gene also reduce the high CpG content of the prokaryotic coding sequence, thereby reducing the chances of epigenetic silencing in mammals.This AAV1 stock expresses an improved Cre (iCre) along with eGFP under a control of the same CMV promoter. The Cre and eGFP was separated by a 2A peptides.

Mammalian homologs of the Drosophila Mad gene include Smad1 (also designated Madr1 or JV4-1), Smad2 (also designated Madr2 or JV18-1), Smad3, Smad4 (also designated DPC4), Smad5, Smad6, Smad7 and Smad8 (also designated Smad9). Smad1 and Smad5 are effectors of BMP2 and BMP4 function while Smad2 and Smad3 are involved in TGFb and activin-mediated growth modulation. Smad4 is required for all of the above activities and necessitates FAST-1 for DNA binding. Smad6 and Smad7 regulate the response to activin/ TGF-beta signaling by interfering with TGF-beta-mediated phosphorylation of other Smad family members.

In the intact cell, DNA closely associates with histones and other nuclear proteins to form chromatin. The remodeling of chromatin is a critical process in transcriptional regulation, and a major source of this remodeling is brought about by the acetylation of nucleosomal histones. Acetylation of lysine residues in the N-terminal tail domain of histone results in an allosteric change in the nucleosomal conformation and an increased accessibility of transcription factors to DNA. Conversely, the deacetylation of histones is associated with transcriptional silencing. Mammalian nuclear histone acetylases include GCN5, PCAF (p300/CBP-associated factor), HAT1, MOF, HBO, MOZ, MORF and TIP60.1.The protein encoded by this gene belongs to class I of the histone deacetylase/acuc/apha family. It has histone deacetylase activity and represses transcription when tethered to a promoter.
