The Quick-RNA Midiprep Kit is designed for the easy, reliable, and rapid isolation of up to 1 mg total RNA from cultured cells or tissue samples. The procedure combines a unique, single-step RNA extraction/binding buffer with Clean-Spin™column technology to yield high quality RNA in about 10 minutes. The Quick-RNA™ Midiprep Kit allows for the efficient recovery of total RNA from 103 to 108 cells or tissue. The method is easy: simply add the provided ZR RNA Buffer to extract total RNA from the cells of interest then purify the RNA using the provided Zymo-Spin™ V-E Columns. The result is highly-concentrated, purified RNA that is suitable for subsequent RNA-based methods including RT-PCR, hybridization, etc.

The CRISPR activation (CRISPRa) system is a variation of the canonical CRISPR-Cas9 used for creation of double-strand breaks in genomic DNA. Zyagen Total RNA is routinely extracted from freshly harvested normal healthy tissues of single donor using classical guanidine isothiocyanate–phenol:chloroform extraction method which allows the rapid isolation of total RNA including microRNAs. RNA is treated with RNase-free DNase to remove residual DNA, precisely quantified, and stored at -80oC. The integrity of each RNA sample as indicated by intact ribosomal RNA is verified by denatured agarose gel electrophoresis. The purity of RNA is assessed by NanoDrop (A260/A280: 1.9-2.1). Residual DNA contamination is tested by PCR.
