GloCell™ Fixable Viability Dye Red 780 is a fluorescent cell viability dye for staining live/dead mammalian cells in applications such as flow cytometry. The dye irreversibly binds intracellular and cell surface amine groups and can be utilized prior to fixation, permeabilization, or cryopreservation. Cells with compromised plasma membranes become permeable to the GloCell™ dye, resulting in a greater fluorescence compared to live cells in a cell viability assay.GloCell™ Fixable Viability Dye Red 780 is excited by the red laser at 633 nm and it has a fluorescence emission maximum of 780 nm that can be detected using the 780/60 band pass filter used to detect APC-Cy7.

Recognizes a single chain glycoprotein of 105/120 kDa, identified as CD30/Ki-1. CD30 is synthesized as a 90 kDa precursor, which is processed in the Golgi complex into a membrane-bound phosphorylated mature 105/120 kDa glycoprotein. In Hodgkin's disease, CD30/Ki-1 antigen is expressed by mononuclear-Hodgkin and multinucleated Reed-Sternberg cells. It is also expressed by the tumor cells of a majority of anaplastic large cell lymphomas as well as by a varying proportion of activated T and B cells. This MAb distinguishes large cell lymphomas derived from activated lymphoid cells from histiocytic malignancies and lymphomas derived from resting and precursor lymphoid cells or from anaplastic carcinomas. Recognizes a 67 kDa glycoprotein, which is identified as CD33. It is a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It belongs to the immunoreceptor tyrosine-based inhibitory motif (ITIM)-containing molecules able of recruiting protein tyrosine phosphatases SHP-1 and SHP-2 to signal assemblies; these ITIMs are also used for ubiquitin-mediated removal of the receptor from the cell surface. CD33 is expressed on cells of myelomonocytic lineage, binds sialic acid residues in N- and O-glycans on cell surfaces, and is a therapeutic target for acute myeloid leukemia. CD33 is expressed on myeloid progenitors, monocytes, granulocytes, dendritic cells and mast cells. Recognizes a protein of 55 kDa, identified as CD25. It is expressed on activated T- and B-cells and activated monocytes/macrophages. With respect to lymphomas, CD25 is present on malignant cells of Hodgkin's disease, HTLV-1 adult T-cell leukemia, cutaneous T-cell lymphoma, and hair cell leukemia. Increased levels of soluble CD25 are observed in the leukemias/lymphomas and inflammatory/ autoimmune diseases. CD25 molecule alone appears to function as a low affinity receptor and associates with CD122 (IL-2R beta chain, p75) and CD132 (common gamma chain) to form the high affinity IL-2 receptor complex. CD25 antibodies detect three epitope regions, A, B and C. This antibody recognizes a protein of 66 kDa-170 kDa, identified as involucrin. In Western blotting of cultured human keratinocytes, this MAb reacts with a 120 kDa protein. It stains the involucrin in a variety of sizes: 170 kDa in MCF-7 cells, a doublet of ~115 kDa and 150 kDa in gorilla and owl monkey, 66 kDa in dog, and a doublet of 105 kDa in pig. Its epitope maps between codon 421-568 of human involucrin. Involucrin is expressed in a range of stratified squamous epithelia, including the cornea, which lacks a distinct cornified layer. In normal epidermis, it is first expressed in the upper spinous layers, and in keratinocyte cultures, all cells that have left the basal layer express it. Recognizes an oncofetal antigen of 220 kDa, identified as a tumor-associated glycoprotein (TAG-72) with properties of a mucin. This MAb defines the mucin-carried sialylated-Tn epitope. TAG-72 is usually expressed by adenocarcinomas, but is negative in mesotheliomas. Studies have reported that this antibody has 80% sensitivity and 93% specificity for pulmonary adenocarcinoma. Therefore, TAG-72 is a useful marker to distinguish between mesothelioma and adenocarcinoma. However, false positive reactions can occur so results must be interpreted with the utmost caution. This antibody may be useful in the differentiation of non-small cell carcinomas from small cell carcinomas of the lung. CD45R, also designated CD45 and PTPRC, has been identified as a transmembrane glycoprotein, broadly expressed among hematopoietic cells. Multiple isoforms of CD45R are distributed throughout the immune system according to cell type. These isoforms arise because of alternative splicing of exons 4, 5, and 6. The corresponding protein domains are characterized by the binding of monoclonal antibodies specific for CD45RA (exon 4), CD45RB (exon 5), CD45RC (exon 6) and CD45RO (exons 4 to 6 spliced out). The variation in these isoforms is localized to the extracellular domain of CD45R, while the intracellular domain is conserved. CD106 (also known as vascular cell adhesion molecule-1, or VCAM-1) is a member of the Ig superfamily of adhesion molecules. It is expressed at high levels on cytokine stimulated vascular endothelial cells, and at minimal levels on un-stimulated endothelial cells. It is also present on follicular and inter-follicular dendritic cells of lymph nodes, myoblasts, and some macrophages. CD106 serves as a ligand for leukocyte beta-1 integrin VLA-4 and mediates cell adhesion of leukocytes to activated endothelium.
