IRDye infrared dyes from LI-COR Biosciences are available for researchers developing applications using near-infrared (NIR) fluorescence. These unique dyes are used in a variety of NIR imaging applications including Western blotting, plate-based assays, protein arrays, tissue section imaging, and molecular activity measurements. Standard NHS ester chemistry is used to produce custom probes labeled with LI-COR IRDye infrared dyes. The NHS ester reactive group provides the functionality for labeling primary and secondary amines, such as lysine residues in proteins.

Dojindo Molecular Technologies Inc DAPGreen–Autophagy Detection(5 nmol)
DAPGreen is used to detect autophagy in live cells. Autophagy is an intracellular degradation system, where dysfunctional proteins and organelles are degraded. In this process, aggregated dysfunctional proteins are surrounded by the double membrane to form an autophagosome. The small fluorescent molecule DAPGreen is used to detect autophagosomes and autolysosomes. The mechanism has been suggested to be that the dye is incorporated into the autophagosome during double-membrane formation via structural features and then emits fluorescence under hydrophobic conditions (λex: 450 nm, λem: 535 nm). DALGreen is cell-permeable, has no requirement of transfection method, and enables live-cell imaging with fluorescence microscopy, plate reader, and flow cytometry. For monitoring only autolysosomes, we recommend DALGreen [D675], as it allows the detection of phagosome-lysosome fusion. Dojindo offers various reagents for autophagy research. Vector Laboratories Wheat Germ Agglutinin (WGA) Fluorescein-labeled 5 mg
Wheat germ agglutinin (WGA) contains a group of closely related isolectins that bind N-acetylglucosamine (preferentially the dimers and trimers of this sugar) and oligosaccharides containing terminal N-acetylglucosamine or chitobiose, structures common to many serum and membrane glycoproteins. Bacterial cell wall peptidoglycans, chitin, cartilage glycosaminoglycans, and glycolipids can also bind WGA. Native WGA also interacts with some glycoproteins via sialic acid residues (see succinylated WGA). This fluorescein-labeled WGA features a ratio of fluorophores to lectin protein that provides optimal staining (excitation 495 nm, emission 515 nm). Supplied as a solution essentially free of unconjugated fluorophores, it is preserved with sodium azide. The recommended inhibiting/eluting sugar is chitin hydrolysate or 500 mM N-acetylglucosamine with salt and/or acid elution.

AEC is a widely used chromogen for immunohistochemical procedures. In the past, AEC chromogens required a water-soluble mounting media because it was soluble in organic solvents. Romulin AEC from Biocare produces a reddish-orange stain. However, it is not soluble in alcohol or xylene-and can be coverslipped just like DAB. It does not fade with permanent mounting media such as PermountTM. When in the presence of a peroxidase enzyme, Romulin AEC produces a reddish-orange stain. This product comes in a 4-component system consisting of ready-to-use buffer, stabilizer, chromogen and hydrogen peroxide.

Chromeo™ P503 detects proteins and peptides by exhibiting a color change from blue to red. After binding to primary amines, it undergos a large shortwave spectral shift of more than 100nm that is visible to the naked eye. There is also a large increase in quantum yield that changes the non- or only weakly fluorescent dye into a bright fluorescent conjugate. The shift of the absorption & emission bands together with the increased fluorescence quantum yield effectively eliminates background from any unbound dye. In addition, because unbound the pyrylium dye is hydrolyzed during the labeling procedure, there is no need for purification following the simple one-step, room-temperature incubation. The labeled protein is ready to use immediately, and any possible background from the unbound dye is eliminated. Due to the unique conjugation mechanism, proteins labeled with Py-Dyes maintain their native charge and isoelectric point.

AdipoGen Biotin Sulfone

Biotin sulfone is an oxidized form of biotin and is as efficient as biotin in streptavidin binding and it binds to avidin with a binding ratio of 0.332 compared with biotin. Biotinylated synthetic oligosaccharides are an attractive tool, as streptavidin coated plates and beads are commercially available and widely use in serological assays. As biotin is chemically reactive, and not compatible with carbohydrate protecting group manipulation, the use of biotin sulfone tagged oligomannosides BSTO for antigen immobilization in candidiasis or allergy diagnosis has been used. The use of biotin sulfone also offers a convenient solution for both oligosaccharide synthesis and immobilization on microspheres and surface plasmon resonance sensors.
