This MAb recognizes the 56.5 kDa (CK10); 50 kDa (CK14); 50 kDa (CK15); 48 kDa (CK16); 40 kDa (CK19) keratins of the acidic (Type I or LMW) subfamily. Twenty human keratins are resolved with two-dimensional gel electrophoresis into acidic (pI 6.0) subfamilies. The acidic keratins have molecular weights (MW) of 56.5, 55, 51, 50, 50', 48, 46, 45, and 40 kDa. MAb AE3 recognizes the 65-67, 64, 59, 58, 56, and 52 kDa keratins of basic subfamily. Many studies have shown the usefulness of keratins as markers in cancer research and tumor diagnosis. Biotium CD53(63-5A3), CF488A conjugate, 0.1mg/mL
Recognizes a protein of 33-55 kDa, identified as CD53 (Workshop V; Code CD53.1). CD53 is expressed on monocytes, and macrophages, granulocytes, dendritic cells, osteoblasts and osteoclasts, NK cells, and on T- and B-cells from every stage of differentiation but is absent from platelets, erythrocytes, and non-haemopoietic cells. CD53 is a member of a family of tetraspan transmembrane proteins, including CD9, CD37, CD63, CD81, and CD82. It associates with integrins, MHC class II molecules, and a tyrosine phosphatase and plays a role in cellular activation as part of a signal transduction complex involving other membrane glycoproteins. Defects of CD53 expression on neutrophils appear to be related with recurrent infectious diseases. CD8 is a cell surface receptor expressed either as a heterodimer with the CD8 beta chain (CD8 alpha/beta) or as a homodimer (CD8 alpha/alpha). A majority of thymocytes and a subpopulation of mature T cells and NK cell express CD8a. CD8 binds to MHC class 1 and through its association with protein tyrosine kinase p56lck plays a role in T cell development and activation of mature T cells. For mature T-cells, CD4 and CD8 are mutually exclusive, so anti-CD8, generally used in conjunction with anti-CD4. It is a useful marker for distinguishing helper/inducer T-lymphocytes, and most peripheral T-cell lymphomas are CD4 /CD8-. Anaplastic large cell lymphoma is usually CD4 and CD8-, and in T-lymphoblastic lymphoma/leukemia, CD4 and CD8 are often co-expressed. This MAb is specific for progesterone. It exhibits minimal cross reactivity with related compounds in ELISA. It reacts with Progesterone-11a-HMS-BSA: 100%; 5-beta-Pregnane-3,20-dione: 48%; 5-alpha-Pregnane-3,20-dione: 26.4%; 17-alpha-Hydroxyprogesterone: 2.5%; 20-alpha-Hydroxyprogesterone: 0.04%. Progesterone is a steroid hormone synthesized from the cholesterol derivative, pregnenolone, in the cortex of the adrenal gland. Progesterone is secreted by the corpus luteum and acts to prepare the endometrium for the implantation of a fertilized egg. Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Biotium Cytokeratin 18(C-04), CF488A conjugate, 0.1mg/mL
This MAb reacts with a wide variety of simple epithelia. It does not react with stratified squamous epithelia. It reacts with epithelial tumors of the gastrointestinal tract, lung, breast, pancreas, ovary, and thyroid. Cytokeratin 18, which belongs to the type A (acidic) subfamily of low molecular weight keratins, exists in combination with cytokeratin 8. It was reported that tissues from gastrointestinal tract are positive for both cytokeratin 8 and 18 but do not contain cytokeratin 14. Biotium p40(Rabbit PAb), CF488A conjugate, 0.1mg/mL
p40 (p63 delta) is a marker determined to be highly specific for squamous basal cells by immunohistochemistry (IHC). The current more routinely recommended marker, p63, appears to have less specificity compared to p40, especially on squamous cell tumors. The ability to differentiate between lung adenocarcinoma vs. squamous cell carcinoma is difficult and has bearing on the different therapeutic avenues for each subtype treatment. p63 antibody's ability to distinguish between the tumor types appears to be inferior when compared to p40. CD31 (PECAM-1) is a transmembrane glycoprotein member of the immunoglobulin supergene family of adhesion molecules. CD31 is expressed by stem cells of the hematopoietic system and is primarily used to identify and concentrate these cells for experimental studies as well as for bone marrow transplantation. Anti-CD31 has shown to be highly specific and sensitive for vascular endothelial cells. Staining of nonvascular tumors (excluding hematopoietic neoplasms) is rare. CD31 MAb reacts with normal, benign, and malignant endothelial cells which make up blood vessel lining. This MAb reacts with a 68 kDa protein, identified as light sub-unit of neurofilaments (NF-L). Neurofilaments make up the main structural elements of axons and dendrites and are found in neurons, peripheral nerves, and sympathetic ganglion cells. Neurofilaments consist of three major subunits with molecular weights of 68 kDa (NF-L), 160 kDa (NF-M) and 200 kDa (NF-H). Anti-neurofilament stains a number of neural, neuroendocrine, and endocrine tumors. Neuromas, ganglioneuromas, gangliogliomas, ganglioneuroblastomas, and neuroblastomas stain positively for anti-neurofilament. Neurofilaments are also present in paragangliomas as well as adrenal and extra-adrenal pheochromocytomas. CD45R, also designated CD45 and PTPRC, has been identified as a transmembrane glycoprotein, broadly expressed among hematopoietic cells. Multiple isoforms of CD45R are distributed throughout the immune system according to cell type. These isoforms arise because of alternative splicing of exons 4, 5, and 6. The corresponding protein domains are characterized by the binding of monoclonal antibodies specific for CD45RA (exon 4), CD45RB (exon 5), CD45RC (exon 6) and CD45RO (exons 4 to 6 spliced out). The variation in these isoforms is localized to the extracellular domain of CD45R, while the intracellular domain is conserved. CD45R functions as a phosphor-tyrosine phosphatase. This MAb reacts with all isoforms of CD45R expressed by all hematopoietic cells, except erythrocytes, having a higher level of expression on lymphocytes than on granulocytes (Workshop IV). Recognizes a protein of 30-33 kDa, which is identified as CD20 (Workshop V; Code CD20.12. Workshop IV; Code B17). It recognizes an extracellular domain of CD20. It is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells. The protein passes through the membrane 4 times with both ends in cytoplasm and exposes one short and one longer loop to the external environment. CD20 is not glycosylated in resting B-cells and its cytoplasmic domains are differentially phosphorylated upon activation. CD10 (also known as Common Acute Lymphocytic Leukemia Antigen (CALLA)) is a type II transmembrane protein. It is a cell surface enzyme with neutral metalloendopeptidase activity, which inactivates a variety of biologically active peptides. CD10 is expressed on the cells of lymphoblastic, Burkitt's, and follicular germinal center lymphomas, and on cells from patients with chronic myelocytic leukemia (CML). Recognizes a protein of 42-46 kDa, identified as MAGE-1. This MAb does not cross-react with MAGE-2, -3, -4, -6 -9, -10, -or -12 protein. Human malignant neoplasms carry rejection antigens that are recognized by the patients' autologous, tumor directed and specific, cytolytic, CD8 T lymphocyte clones (CTL). The MAGE family of genes codes an important group of antigens. It was identified that melanomas and primary glial brain tumors express common melanoma associated antigens (MAAs).
