Strep-Tactin TACS Agarose contains agarose macrobeads coated with the streptavidin variant Strep-Tactin. A special crosslinking permits the use for cell isolation and depletion approaches. It was developed for column-based Traceless Affinty Cell Selection (Fab-TACS/Nano-TACS). In this approach, Fab fragments fused to a Twin-Strep-tag (Fab-Streps) or nanobodies (Nano-Streps) are bound to Strep-Tactin on the agarose matrix. After specifically capturing cells, Fab-/Nano-Streps are released from the Strep-Tactin Agarose by biotin addition. In this way, the eluted cells are ready for further downstream applications. The same principle is applicable for exosome isolation (Fab-TACS for exosomes). Alternatively to Fab-Streps/Nano-Streps, it is possible to bind Twin-Strep-tagged, desthiobiotinylated or biotinylated antibodies (irreversible binding) to this resin. This makes Strep-Tactin TACS Agarose also suitable for negative cell selection or depletion approaches.
