A Deep Dive into BD Horizon Real Dye Spillover & Spectral Flow
Spillover spreading is the biggest constraint on high-parameter panel design. When a fluorochrome is excited by multiple lasers, its broad emission profile interferes with neighboring channels, forcing complex compensation adjustments that degrade resolution. PE and PerCP tandems are the most common offenders, excited simultaneously by both blue and yellow-green laser lines.
BD Horizon Real Dyes are engineered to solve this. Each dye is laser-specific, dramatically reducing cross-laser excitation compared to PE and PerCP tandem dyes. As shown in Figure 1, dyes such as PerCP Cy5.5, Bio-Rad StarBright™ Blue 700, and BD Horizon Brilliant™ Blue 700 produce multi peak emission profiles across multiple laser lines, while 705 produces a clean single-peak profile with minimal cross-channel interference.
Optimized for Spectral Flow Cytometry
The laser-specific design makes them particularly powerful on spectral flow cytometers, where unique emission signatures are essential for accurate unmixing. RB780 and RY775 show reduced cross-laser excitation compared to PE-Cy7, enabling superior performance on instruments with blue and yellow-green lasers, such as five-laser BD FACSymphony™ Analyzers and spectral flow cytometers, reducing spectral spread and increasing panel design flexibility (Figure 2).

Resolution: High stain index values across all four laser lines, with brightness levels matching each dye to the right marker, pairing brighter options with dimly expressed targets to reduce spillover spread across the panel.
Photostability: Consistent MFI and spectral profiles even after extended light exposure, reducing compensation or unmixing errors between reference controls and samples.
Intracellular Staining: Supports surface and intracellular targets including cytokines, transcription factors, cell cycle markers and phosphoproteins, using standard fixation and permeabilization systems.
Buffer Compatibility: Broadly compatible with BD Cytofix™, BD Phosflow™ Perm Buffer III, BD Cytofix/Cytoperm™, BD FACS™ Permeabilizing Solution 2, and other common systems, where PE and PerCP tandems are prone to signal degradation.
Monocyte Background: Engineered to minimize nonspecific monocyte binding, a chronic issue with dyes like PE-Cy7. BD Horizon Real Dyes deliver cleaner staining profiles and fewer false-positive signals in whole blood panels.
