VS-5584 is a pan-PI3K/mTOR kinase inhibitor that simultaneously blocks both mTORC2 and mTORC1. This ATP-competitive inhibitor selectively targets PI3K/mTOR signaling with low nanomolar potency against all human Class I PI3K isoforms and mTOR kinase. It preferentially inhibits cancer stem cells in HMLE breast cancer cells, reducing Aldefluor-positive cancer stem cells. In vivo, it reduces cancer stem cells in breast cancer tumors and induces tumor regression in Docetaxel-resistant models. Rapidly absorbed with a tmax of 0.9 hours and an elimination half-life of 10 hours, it leads to significant tumor growth inhibition.
