4-Methylumbelliferone (4-MU) is a small-molecule inhibitor of hyaluronic acid (HA) synthesis commonly employed in biomedical research Excessive HA production often resulting from the activation of HA synthase 2 (HAS2) correlates with increased cell migration invasion and metastatic behavior in various tumor cell types Mechanistically 4-MU suppresses HA biosynthesis primarily through activation of neutral sphingomyelinase 2 (NSMase2) elevating intracellular ceramide levels Elevated ceramide subsequently stimulates the activity of protein phosphatase PP2A leading to reduced phosphorylation of Akt decreased HAS2 activity and inhibition of HA generation Additionally ceramide elevation induced by 4-MU promotes activation of p53 and caspase-3 signaling pathways alongside downregulation of SIRT1 collectively contributing to reduced tumor cell viability migration and metastasis in vitro Clinical studies investigating the therapeutic potential of 4-MU are ongoing
