CQ211 is a potent and selective RIOK2 inhibitor with a Kd of 6.1 nM. It exhibits potent proliferation inhibition activity against multiple cancer cell lines. In vitro, CQ211 suppresses the phosphorylation of mTOR in cancer cells. It effectively inhibits the proliferation of MKN-1 and HT-29 cells, with IC50 values of 0.61 μM and 0.38 μM, respectively. In vivo, administration of CQ211 at 25 mg/kg was observed to inhibit the MKN-1 xenograft mouse model, resulting in a tumor growth inhibition (TGI) of 30.9%.
