A-395 hydrochloride is a small-molecule chemical probe that antagonizes the EED subunit of the Polycomb repressive complex 2 (PRC2). It binds in the H3K27me3-binding pocket of EED, preventing allosteric activation of EZH2 and reducing H3K27 methylation; it shows potent inhibition of the PRC2 complex (reported IC50 ≈ 18 nM). The compound is supplied as the hydrochloride salt and is commonly used in biochemical and cellular studies.
- Potent EED antagonist with low-nanomolar PRC2 inhibition.
- Prevents allosteric activation of EZH2 and reduces H3K27 methylation.
- Validated in structural and biochemical studies for target engagement.
- Suitable as a chemical probe for studying PRC2 biology in cells and assays.
- Supplied as a hydrochloride salt with good solubility in DMSO.
- Available in solution formats for direct use in biochemical assays.
