This gene encodes a 190 kDa nuclear phosphoprotein that helps maintain genomic stability and functions as a tumor suppressor. It spans 22 exons over ~110 kb and forms part of the BRCA1-associated genome surveillance complex (BASC), interacting with DNA repair proteins and RNA polymerase II. Through its C-terminal domain, it also associates with histone deacetylase complexes, contributing to transcription regulation, DNA repair, and recombination. Mutations in BRCA1 account for ~40% of inherited breast cancers and over 80% of inherited breast and ovarian cancers. Alternative splicing affects its localization and function, with many transcript variants described, some linked to disease. A related pseudogene also exists on chromosome 17.
