This gene encodes a TNF-receptor superfamily member with a death domain that regulates programmed cell death and is linked to immune disorders and cancers. Upon ligand binding, it forms a death-inducing signaling complex with FADD, caspase 8, and caspase 10, triggering a caspase cascade and apoptosis. It can also activate NF-kappaB, MAPK3/ERK1, and MAPK8/JNK pathways, contributing to cell proliferation in fibroblasts and T cells. Multiple transcript variants exist, some subject to nonsense-mediated decay. Isoforms lacking the transmembrane domain may inhibit apoptosis mediated by the full-length receptor.
