Hyaluronan (HA) is a high molecular weight polysaccharide found in the extracellular matrix, composed of alternating glucuronic acid and N-acetylglucosamine linked by β-1,3 and β-1,4 bonds. Synthesized by membrane-bound synthases, HA is extruded into the extracellular space and plays roles in joint lubrication, space filling, and supporting cell migration. It is actively produced during wound healing and tissue repair and is involved in leukocyte homing via interaction with CD44. HA levels are altered in conditions like rheumatoid arthritis, and overexpression of HA receptors is linked to tumor metastasis. HAS1 encodes a hyaluronan synthase and shares homology with similar genes in other species. Alternative splicing produces multiple transcript variants.
