This gene encodes a preproprotein processed into alpha and beta chains that form haptoglobin, a tetramer that binds free plasma hemoglobin. This binding prevents kidney damage and iron loss while enabling hemoglobin degradation. Mutations in this gene or its regulatory regions cause ahaptoglobinemia or hypohaptoglobinemia. It is also linked to diabetic nephropathy, coronary artery disease in type 1 diabetes, Crohns disease, inflammatory disorders, primary sclerosing cholangitis, susceptibility to Parkinson’s disease, and reduced risk of Plasmodium falciparum malaria. The protein also shows antimicrobial activity. A similar duplicated gene lies nearby on chromosome 16, and multiple transcript variants encoding different isoforms exist.
