This gene encodes a C-type lectin involved in cell adhesion and pathogen recognition. It recognizes a variety of pathogens, including mycobacteria (leprosy, tuberculosis), viruses (Ebola, hepatitis C, HIV-1, Dengue, SARS-CoV), and more. The protein has four domains: a C-terminal carbohydrate recognition domain, a flexible neck domain, a transmembrane region, and an N-terminal cytoplasmic domain for internalization. It is related to the CLEC4M gene (L-SIGN), differing in viral recognition and expression patterns. This gene is highly expressed on dendritic cells. Polymorphisms in the neck region are linked to HIV-1 protection, and promoter SNPs are associated with resistance and susceptibility to diseases, including rs4804803, which correlates with SARS severity.
