Myosin phosphatase target subunit 1 (MBS), also known as the myosin-binding subunit, is part of myosin phosphatase, which regulates actin-myosin interaction through Rho GTPases. RhoA, in its active GTP-bound form, interacts with MBS to control myosin light chain (MLC) phosphorylation, affecting smooth muscle contraction and actin-myosin interaction in nonmuscle cells. Rho-kinase, activated by RhoA, phosphorylates MBS, inhibiting myosin phosphatase. Overexpression of RhoA increases MBS and MLC phosphorylation. This protein plays a key role in regulating actin dynamics. Multiple transcript variants encoding distinct isoforms have been identified.
