Atuveciclib is a potent and highly selective oral PTEFb/CDK9 inhibitor that targets CDK9/CycT1 with an IC50 of 13 nM. It demonstrates potent antiproliferative activity against various cell lines, including HeLa (IC50=920 nM) and MOLM-13 (IC50=310 nM) in vitro. In vivo studies in the MOLM-13 xenograft model show high antitumor efficacy with dose-dependent results and good tolerability, with less than 10% mean body weight reduction. It also exhibits low blood clearance in rats.
