AZD5582 dihydrochloride is an antagonist of the inhibitor of apoptosis proteins (IAPs). It binds to the BIR3 domains of cIAP1, cIAP2, and XIAP with IC50s of 15, 21, and 15 nM, respectively, and induces apoptosis. This compound has demonstrated a range of biological activities both in vitro and in vivo.
- Inhibits cell viability in H1975 NSCLC cells when cooperating with IFNγ or viral dsRNA.
- Downregulates cIAP-1, activates RIPK1, and triggers activation of extrinsic and intrinsic apoptosis pathways in H1975 NSCLC cells.
- Induces apoptosis and active caspase-3/8 activities when co-treated with IFNγ in HCC827 NSCLC cells.
- Causes degradation of cIAP1 and caspase 3 cleavage in tumor cells of MDA-MB-231 xenograft models.
- Achieves substantial tumor regressions in MDA-MB-231 xenograft models with two weekly doses.
