Azido-PEG4-hydrazide-Boc is a PEG-based PROTAC linker used in the synthesis of PROTACs. It functions as a click chemistry reagent containing an Azide group, enabling it to participate in copper-catalyzed azide-alkyne cycloaddition reactions (CuAAc) with molecules having Alkyne groups. It can also undergo strain-promoted alkyne-azide cycloaddition (SPAAC) reactions with molecules containing DBCO or BCN groups. PROTACs exploit the intracellular ubiquitin-proteasome system to degrade target proteins, consisting of two different ligands connected by a linker: one for an E3 ubiquitin ligase and one for the target protein.
