CLP257 is a selective K+-Cl cotransporter KCC2 activator with an EC50 of 616 nM. It does not act on NKCC1, GABAA receptors, KCC1, KCC3, or KCC4. This compound helps restore impaired chloride transport in neurons where KCC2 activity is reduced. It has also been shown to alleviate hypersensitivity in rats experiencing neuropathic pain. CLP257 also influences plasmalemmal KCC2 protein turnover after translation.
