Coelenterazine h (2-(4-Dehydroxy) coelenterazine) is a synthetic derivative of coelenterazine. The luminescence intensity of its aequorin complex is more than 10 times higher than that of native coelenterazine. Coelenterazines are luminescent enzyme substrates for apoaequorin and Renilla luciferase. Apoaequorin is used as a calcium indicator. It binds coelenterazine to form an aequorin complex that emits light upon calcium binding and measures concentrations ~0.1 uM to >100 uM. Renilla luciferase is widely used as a reporter of transcription regulation and as a donor in bioluminescence resonance energy transfer (BRET) to study protein-protein interactions. Coelenterazines are also used for chemiluminescent detection of superoxide anions and peroxynitrite in cells or tissues.
