CMPD-39 is a highly selective, non-covalent inhibitor of the ubiquitin-specific protease USP30, with an IC50 of approximately 20 nM. It shows significant selectivity over other deubiquitinase family members. By inhibiting USP30's deubiquitinating activity, CMPD-39 increases ubiquitination of mitochondrial proteins (TOMM20 and SYNJ2BP), promoting phosphoubiquitin accumulation. This accelerates both mitochondrial autophagy (mitophagy) and peroxisomal autophagy (pexophagy).
