This gene encodes coagulation factor III, a cell surface glycoprotein that initiates blood coagulation cascades. It acts as the high-affinity receptor for coagulation factor VII, forming a complex that triggers the coagulation protease cascades through limited proteolysis. Unlike other cofactors, factor III is fully functional when expressed on cell surfaces, such as on monocytes. It has three domains: extracellular, transmembrane, and cytoplasmic. Platelets and monocytes express this factor under procoagulatory and proinflammatory stimuli. It plays a significant role in HIV-associated coagulopathy and has been linked to COVID-19 severity and mortality through platelet-dependent monocyte expression. Factor III is the only protein in the coagulation pathway without a known congenital deficiency. Alternative splicing generates multiple transcript variants.
