Congo Red Agar is a differential microbiological medium used for detecting biofilm formation and slime production in bacteria, particularly Staphylococcus species. Each sterile petri dish contains 30mL of a nutrient rich agar base supplemented with Congo Red dye and sucrose. The increased fill volume helps maintain moisture and nutrient stability during extended incubation periods that may last up to 72 hours. Microorganisms capable of producing extracellular polysaccharides bind the dye and develop dark, almost black colonies with a dry or crusty appearance. Non slime producing strains typically form smooth pink or red colonies. This visual contrast allows researchers to rapidly distinguish biofilm producing organisms, making the medium valuable in clinical microbiology, biofilm research, and studies involving bacterial virulence and adhesion behavior.
