Ecopladib is a sub-micromolar inhibitor of cytosolic phospholipase A2α (cPLA2α), with IC50s of 0.15 μM in GLU micelle and 0.11 μM in rat whole blood assays. It inhibits cPLA2α in PAPE liposome assay at 73% at 37 nM and is inactive against COX-1 and COX-2. It also inhibits the production of prostaglandins and leukotrienes. In vivo, Ecopladib is orally efficacious for inhibiting COX-2 derived PGE2 formation and reducing carrageenan-induced paw swelling.
