Ensitrelvir fumarate (S-217622) is the first orally active non-covalent, non-peptidic SARS-CoV-2 3CL protease inhibitor with an IC50 of 13 nM. It demonstrates EC50 values of approximately 0.4 μM against wild-type SARS-CoV-2 and its Alpha, Beta, Gamma, and Delta variants in cytopathic effect (cpe)-inhibition assays. This compound also dose-dependently inhibits the intrapulmonary replication of SARS-CoV-2 in mice and shows activity against SARS-CoV and MERS-CoV.
