FKBP12 PROTAC RC32 is a potent FKBP12 degrader based on PROTAC technology. This degrader combines Rapamycin and a Cereblon E3 ubiquitin ligase ligand, Pomalidomide. It achieves approximately 50% protein degradation (DC50) of FKBP12 at 0.3 nM in Jurkat cells within 12 hours. In vivo, it effectively degrades FKBP12 in most organs of treated mice, Bama pigs, and rhesus monkeys, with oral administration also showing significant degradation in mice.
