A mouse embryo fibroblast cell line engineered to package ecotropic retroviral genomes and serve as a transfection host for production of retroviral vectors for delivery of foreign genes into susceptible eukaryotic cells. Derived from NIH 3T3 cells via electroporation of plasmids encoding gag/pol and env, it is commonly used for retroviral vector production and transfection workflows.
- Suitable for packaging psi-containing nucleic acids into ecotropic retrovirus genomes.
- Adherent growth with fibroblast morphology.
- Recommended culture at 37°C in 95% air, 5% CO2.
- Complete medium: DMEM supplemented with hypoxanthine (0.015 mg/mL), xanthine (0.25 mg/mL), mycophenolic acid (0.025 mg/mL), and 90% calf serum.
- Subculture ratio typically 1:5 to 1:10; renew medium twice weekly.
- Store cryogenically in vapor phase of liquid nitrogen; cryopreserve in medium with 5% DMSO.
