It utilizes a monoclonal antibody specific for CD146(MCAM / MUC18) coated on a 96-well plate. Standards and samples are added tothe wells, and any CD146 present binds to the immobilized antibody. The wellsare washed and a horseradish peroxidase conjugated rabbit anti-CD146 polyclonalantibody is then added, producing an antibody-antigen-antibody "sandwich". Thewells are again washed and TMB substrate solution is loaded, which producescolor in proportion to the amount of CD146 present in the sample. To end theenzyme reaction, the stop solution is added and absorbances of the microwellare read at 450 nm.
