Ibutilide (U70226E free base) is an action potential-prolonging antiarrhythmic agent that acts as a potent blocker of the rapidly activating delayed rectifier K+ current (IKr) in AT-1 cells. It inhibits Cav1.2 current in CHO cells and prolongs cardiac repolarization in vivo.

• Potent IKr blocker with an EC50 of 20 nM at +20 mV in atrial tumor myocytes.
• Inhibits Cav1.2 current with an IC50 of 62.5 μM in CHO cells.
• Blocks IKr in HERG+MDR1*1 and HERG alone, showing resistance in MDR1*7 expressing cells.
