ICA 110381 is an orally active Kv7.2/Kv7.3 (KCNQ2/3) potassium channel activator with anticonvulsive properties. It acts as a KCNQ2/Q3 activator with an EC50 of 0.38 μM and also as a KCNQ1 antagonist with an IC50 of 15 μM.
- Can be administered orally.
- Targets specific potassium channels.
- Helps in preventing or reducing the severity of seizures.
- Induces rubidium efflux in CHO cells expressing KCNQ2 or KCNQ3 (EC50 = 0.38 μM).
- Inhibits rubidium efflux from CHO cells expressing KCNQ1 (EC50 = 15 μM).
- Damps neuronal excitability in CA1 pyramidal cells by increasing M-currents (IM), reducing input resistance, and shifting the resting membrane potential hyperpolarizing direction.
- Elevates threshold for afterdischarges, reducing seizure severity and duration in amygdala-kindled rats.
