Activate the Activin/BMP/TGF-β pathway with inducer of definitive endoderm 1 (IDE1; Borowiak et al. Cell Stem Cell, 2009). This small molecule induces the differentiation of mouse or human embryonic stem (ES) cells to definitive endoderm in the absence of Activin A, NODAL, or feeder cells (Borowiak et al. Cell Stem Cell, 2009), and the differentiation of mouse or human pluripotent stem cells by activating SMAD2 phosphorylation and NODAL expression (Borowiak et al. Cell Stem Cell, 2009).
