LEQ506 is a second-generation inhibitor of smoothened (Smo) with IC50 values of 2 nM in human and 4 nM in mouse. It inhibits Hedgehog (Hh) signaling in a human cell line (HEPM) by decreasing Gli mRNA, with an IC50 approximately 6-fold lower than that of Compound 2. At 1%, LEQ506 achieves 80% to 90% inhibition for Gli1 and 60% to 70% for Ptch1.
