MDA-T68 (CRL-3353) is a human epithelial-like cell line derived from thyroid tissue of a 75-year-old male with the follicular variant of papillary thyroid carcinoma. It is intended for in vitro research applications such as therapeutic testing and molecular studies; the line harbors HRAS Q61K and TERT promoter -124 C>T mutations and shows detectable TTF-1 expression.
- Adherent growth with epithelial morphology.
- Population doubling time approximately 45 hours.
- Supplied frozen in a 1.0 mL vial with approximately 2.0 x 10^6 cells.
- Recommended medium: RPMI-1640 supplemented with 10% fetal bovine serum, 2 mM L-glutamine, and 0.1 mM nonessential amino acids.
- Culture conditions: 37°C, 95% air/5% CO2; maintain 5 x 10^4 to 2 x 10^5 cells/cm^2.
- Subculture ratio 1:3 to 1:8; renew medium 2-3 times per week.
- Store long term in the vapor phase of liquid nitrogen; handle thawed cultures under aseptic conditions.
